摘要 目的:探讨血清长链非编码核糖核酸(lncRNA)烟酰胺核苷酸反义转氢酶RNA1(NNT-AS1)、微小核糖核酸(miR)-582-5p与支气管哮喘患儿气道炎症、气道高反应的关系。方法:选择120例支气管哮喘患儿,根据相关指南分为缓解期组(n=52)与发作期组(n=68)。另选择同期60例健康儿童作为对照组。对比三组血清lncRNA NNT-AS1、miR-582-5p相对表达量及气道炎症细胞因子[白介素-17(IL-17)、肿瘤坏死因子-α(TNF-α)]水平、气道反应性指标[基础呼吸阻力(Rrs cont)、最小诱发累积剂量(Dmin)、基础呼吸传导率(Grs cont)]。Pearson相关性检验分析血清lncRNA NNT-AS1、miR-582-5p表达与血清气道炎症细胞因子、气道反应性指标的关系。结果:缓解期组、发作期组lncRNA NNT-AS1相对表达量、血清IL-17、TNF-α水平、Rrs cont高于对照组,且发作期组高于缓解期组(P<0.05)。缓解期组、发作期组miR-582-5p相对表达量、Grs cont、Dmin低于对照组,且发作期组低于缓解期组(P<0.05)。支气管哮喘患儿血清lncRNA NNT-AS1表达与IL-7、TNF-α、Rrs cont呈正相关(r>0,P<0.05),与Grs cont、Dmin呈负相关(r<0,P<0.05);血清miR-582-5p表达与IL-17、TNF-α、Rrs cont呈负相关(r<0,P<0.05),与Grs cont、Dmin呈正相关(r>0,P<0.05)。结论:支气管哮喘患儿血清lncRNA NNT-AS1呈高表达,miR-582-5p呈低表达,二者表达与患儿哮喘分期、气道炎症、气道高反应密切相关。 |

ABSTRACT Objective: To investigate the relationship between serum long non-coding RNA (lncRNA) nicotinamide nucleotide antisense transferase RNA1 (NNT-AS1), microRNA (miR)-582-5p and airway inflammation and airway hyperresponsiveness in children with bronchial asthma. Methods: 120 children with bronchial asthma were divided into remission group (n=52) and attack group (n=68) according to the relevant guidelines. Another 60 healthy children in the same period were selected as the control group. The relative expression of serum lncRNA NNT-AS1, miR-582-5p and serum airway inflammatory cytokines [interleukin-17 (IL-17), tumor necrosis factor-α (TNF-α)], airway responsiveness indicators [basic respiratory resistance (Rrs cont), minimum induced cumulative dose (Dmin), basic respiratory conductivity (Grs cont)] were compared among three groups. The relationship between serum lncRNA NNT-AS1, miR-582-5p expression and airway inflammatory cytokines, airway responsiveness indicators were analyzed by Pearson correlation test. Results: The relative expression of lncRNA NNT-AS1, serum levels of IL-17, TNF-α and Rrs cont in remission group and attack group were higher than those in control group, and those in attack group were higher than those in remission group (P<0.05). The relative expression of miR-582-5p, Grs cont and Dmin in remission group and attack group were lower than those in control group, and those in attack group were lower than those in remission group(P<0.05). The expression of serum lncRNA NNT-AS1 in bronchial asthma children was positively correlated with IL-7, TNF-α and Rrs cont (r>0, P<0.05), and negatively correlated with Grs cont and Dmin (r<0, P<0.05). The expression of serum miR-582-5p was negatively correlated with IL-17, TNF-α and Rrs cont (r<0, P<0.05), and positively correlated with Grs cont and Dmin(r>0, P<0.05). Conclusion: Serum lncRNA NNT-AS1 is highly expressed and miR-582-5p is lowly expressed in children with bronchial asthma, the expression of both are closely related to asthma stage, airway inflammation and airway hyperresponsiveness. |
