      摘要 目的:探究2月龄阿尔茨海默症模型小鼠(APP/PS1)初级视皮层(primary visual cortex, V1)神经元在不同视觉刺激模式下的活性变化,解析APP/PS1小鼠早期V1的功能改变。方法:通过免疫荧光染色分析2月龄APP/PS1小鼠初级视皮层内Aβ表达情况,并选取2月龄APP/PS1和野生型(wild type, WT)小鼠,给予其不同条件视觉刺激后,通过c-Fos荧光染色解析V1神经元的活性变化。结果:2月龄APP/PS1小鼠V1内存在Aβ表达但未形成明显淀粉样斑块。在没有外界额外视觉刺激的情况下,2月龄APP/PS1小鼠V1中c-Fos阳性率高于WT小鼠(P<0.01);当给予小鼠外界30分钟的视觉刺激或者视觉刺激后的90分钟,2月龄APP/PS1与WT小鼠V1中c-Fos阳性率均没有明显差异。结论:2月龄APP/PS1小鼠V1神经元在无外界视觉刺激时处于过度活跃状态,其早期视皮层神经元的活动模式可能存在改变。

      ABSTRACT Objective: Investigate the changes of neuronal activity in the primary visual cortex of 2-month-old APP/PS1 mice, an Alzheimer's disease model, under different visual stimulation conditions, and elucidate the early functional alterations in the visual cortex of APP/PS1 mice. Methods: The expression of Aβ in the primary visual cortex of 2-month-old APP/PS1 mice was analyzed by immunofluorescence staining, and 2-month-old APP/PS1 and wild type (WT) mice were selected and subjected to different visual stimulation conditions. The changes of neuronal activity in the primary visual cortex were analyzed by performing immunofluorescent staining. Results: Aβ is expressed in V1 of 2-month-old APP/PS1 mice, but no obvious amyloid plaques has been found. In the absence of external visual stimulation, the c-Fos positive rate in the V1 of 2-month-old APP/PS1 mice is higher than that in wild-type mice(P<0.01). When given 30 minutes of visual stimulation to mice or 90 minutes after visual stimulation, there is no significant difference in c-Fos positive rate in the V1 of 2-month-old APP/PS1 and WT mice in both cases. Conclusion: The neurons in the V1 of 2-month-old APP/PS1 mice are in a hyperactive state in the absence of external visual stimulation, suggesting altered activity patterns of cortical neurons in the visual cortex of young APP/PS1 mice.
