      摘要 目的:探讨血清微小核糖核酸(miR)-191-5p、miR-101-3p与胃癌患者临床病理特征、磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)信号通路及预后的关系。方法: 选取2019年1月至2021年1月我院收治的182例胃癌患者(胃癌组)和同期于体检中心体检健康的90例体检者(对照组)。检测两组血清miR-191-5p、miR-101-3p以及PI3K mRNA、Akt mRNA表达。Pearson法分析血清miR-191-5p、miR-101-3p与PI3K mRNA、Akt mRNA的相关性。分析血清miR-191-5p、miR-101-3p与胃癌患者临床病理特征的关系。Kaplan-Meier曲线分析不同miR-191-5p、miR-101-3p表达胃癌患者的3年总生存(OS)率。结果:胃癌组血清miR-191-5p、miR-101-3p表达低于对照组(P<0.05),PI3K mRNA、Akt mRNA表达高于对照组(P<0.05)。胃癌患者血清miR-191-5p、miR-101-3p表达与PI3K mRNA、Akt mRNA均呈负相关(P<0.05)。血清miR-191-5p、miR-101-3p表达与TNM分期、淋巴结转移、浸润深度、分化程度有关(P<0.05)。miR-191-5p低表达胃癌患者3年OS率低于miR-191-5p高表达胃癌患者,miR-101-3p低表达胃癌患者3年OS率低于miR-101-3p高表达胃癌患者(P<0.05)。结论:胃癌患者血清miR-191-5p、miR-101-3p表达显著下调,与TNM分期、淋巴结转移、浸润深度、分化程度有关,可能通过激活PI3K /Akt 信号通路导致不良预后。

      ABSTRACT Objective: To investigate the relationship between serum microribonucleic acid (miR)-191-5p, miR-101-3p and clinicopathological features, phosphatidylinositol 3 kinase (PI3K)/protein kinase B (Akt) signaling pathway and prognosis in gastric cancer patients. Methods: 182 gastric cancer patients (gastric cancer group) admitted to our hospital from January 2019 to January 2021 and 90 healthy subjects (control group) who underwent physical examination in the physical examination center during the same period were selected. The expressions of serum miR-191-5p, miR-101-3p, PI3K mRNA and Akt mRNA were detected in two groups. The correlation between serum miR-191-5p, miR-101-3p and PI3K mRNA, Akt mRNA was analyzed by Pearson. The relationship between serum miR-191-5p, miR-101-3p and clinicopathological features of gastric cancer patients was analyzed. The 3-year overall survival (OS) rate of gastric cancer patients with different miR-191-5p and miR-101-3p expression was analyzed by Kaplan-Meier curve. Results: The expression of serum miR-191-5p and miR-101-3p in gastric cancer group was lower than that in control group (P<0.05), and the expression of PI3K mRNA and Akt mRNA was higher than that in control group (P<0.05). The expression of serum miR-191-5p and miR-101-3p in gastric cancer patients was negatively correlated with PI3K mRNA and Akt mRNA (P<0.05). The expression of serum miR-191-5p and miR-101-3p was related to TNM stage, lymph node metastasis, depth of invasion and differentiation of gastric cancer (P<0.05). The 3-year OS rate of gastric cancer patients with low expression of miR-191-5p was lower than that of gastric cancer patients with high expression of miR-191-5p, and the 3-year OS rate of gastric cancer patients with low expression of miR-101-3p was lower than that of gastric cancer patients with high expression of miR-101-3p (P<0.05). Conclusion: The expression of serum miR-191-5p and miR-101-3p in gastric cancer patients are significantly down-regulate, which are relate to TNM stage, lymph node metastasis, depth of invasion and degree of differentiation, may lead to poor prognosis by activating PI3K/Akt signaling pathway.
