      摘要 目的:分析黑树莓多酚经调节STAT3/Survivin信号途径对胃癌大鼠的作用机制。方法:选取40只雄性健康SD大鼠,取10只作为空白对照组,另30只制备胃癌大鼠模型,建模成功后分为模型组和黑树莓多酚(低、高剂量)组,各组10只;给予黑树莓多酚(低、高剂量)组大鼠5%、10%黑树莓多酚饲料喂养,给予空白对照组和模型组大鼠给予普通饲料喂养,持续干预8周后测量大鼠体质量和肿瘤体积;并采用Western blot检测STAT3、Survivin蛋白表达水平;采用酶联免疫吸附法(ELISA)检测组织病理学和炎症因子水平。结果:黑树莓多酚(低、高剂量)组体质量、肿瘤体积均低于模型组,高剂量组明显低于低剂量组(P<0.05)。STAT3、Survivin蛋白表达水平比较,黑树莓多酚(低、高剂量)组均低于模型组,高剂量组明显低于低剂量组(P<0.05)。黑树莓多酚(低、高剂量)组胃动素(MTL)、胃蛋白酶原Ⅰ(PGⅠ)、胃泌素(GAS)水平明显高于模型组,高剂量组明显高于低剂量组(P<0.05)。白介素1(IL-1)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平比较,黑树莓多酚(低、高剂量)组显著高于模型组,高剂量组显著高于低剂量组(P<0.05)。结论:黑树莓多酚通过调节STAT3/Survivin信号途径的方式有利于抑制胃癌大鼠肿瘤生长,并有效调节胃肠激素,改善胃功能,同时还能通过影响炎症因子水平的方式干扰胃癌进展。

      ABSTRACT Objective: The mechanism of action of black raspberry polyphenols on gastric cancer rats by regulating STAT3 / Survivin signaling pathway was analyzed. Methods: A total of 40 male healthy SD rats were selected, 10 of which were taken as the blank control group, and the other 30 were used to prepare the gastric cancer rat model. After successful modeling, they were divided into the model group and the black raspberry polyphenols (low and high dose ) group, with 10 rats in each group. The rats in the black raspberry polyphenol (low and high dose) group were fed with 5 % and 10 % black raspberry polyphenol feed, and the rats in the blank control group and the model group were fed with normal feed. After 8 weeks of continuous intervention, the body weight and tumor volume of the rats were measured. The expression levels of STAT3 and Survivin protein were detected by Western blot. Histopathological and inflammatory factor levels were detected by enzyme-linked immunosorbent assay (ELISA). Results: The body weight and tumor volume of the black raspberry polyphenol (low and high dose) groups were lower than those of the model group, and the high dose group was significantly lower than the low dose group(P<0.05). Compared with the expression levels of STAT3 and Survivin protein, the black raspberry polyphenol ( low and high dose ) groups were lower than the model group, and the high dose group was significantly lower than the low dose group (P<0.05). The levels of motilin (MTL), pepsinogen I (PGI) and gastrin (GAS) in black raspberry polyphenol (low and high dose) groups were significantly higher than those in model group, and those in high dose group were significantly higher than those in low dose group (P<0.05). The levels of interleukin-1 (IL-1), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in black raspberry polyphenol (low and high dose) groups were significantly higher than those in model group, and those in high dose group were higher than those in low dose group (P<0.05). Conclusion: Black raspberry polyphenols can inhibit the growth of gastric cancer in rats by regulating STAT3 / Survivin signaling pathway, effectively regulate gastrointestinal hormones, improve gastric function, and interfere with the progression of gastric cancer by affecting the level of inflammatory factors.
