      摘要 目的:探究酪氨酸磷酸酶Z1型受体(PTPRZ1)、环氧合酶-2(COX-2)、人端粒酶逆转录酶(hTERT)在口腔黏膜下纤维性变(oral submucosal fibrosis, OSF)癌变中的表达及相关性。方法:采集2018年6月-2019年6月于我院接受治疗的OSF癌变患者OSF癌变组织30例,作为本文研究OSF癌变组;采集2018年6月-2019年6月于我院接受治疗的OSF患者OSF组织30例,作为本文研究OSF组;同时选取无槟榔、烟酒嗜好、正常口腔粘膜组织30例作为本文研究正常组。Western blot法检测PTPRZ1、hTERT相对表达量,酶联免疫吸附法检测COX-2表达,对待测标本进行免疫组化染色,并对PTPRZ1、COX-2、hTERT在OSF癌变中的相关性进行分析。结果:OSF组PTPRZ1、COX-2、hTERT相对表达量均高于正常组(P<0.05);OSF癌变组PTPRZ1、COX-2、hTERT相对表达量均高于正常组、OSF组(P<0.05)。低分化OSF癌变组织中PTPRZ1、COX-2、hTERT相对表达量均高于高分化、中分化OSF癌变组织(P<0.05)。Ⅳ期OSF癌变组织中PTPRZ1、COX-2、hTERT相对表达量均高于Ⅱ期、Ⅲ期OSF癌变组织(P<0.05)。OSF癌变组织中PTPRZ1、COX-2、hTERT相对表达量呈正相关。结论:PTPRZ1、COX-2、hTERT在OSF癌变中呈异常高表达,低分化、病理分期较高的OSF癌变中PTPRZ1、COX-2、hTERT表达较高,并且PTPRZ1、COX-2、hTERT在OSF癌变中表达呈正相关。

      ABSTRACT Objective: To investigate the expression and correlation of tyrosine phosphatase Z1 receptor (ptprz1), cyclooxygenase-2 (COX-2) and human telomerase reverse transcriptase (hTERT) in oral submucosal fibrocarcinogenesis. Methods: From June 2018 to June 2019, 30 cases of oral submucous fibrotic canceration(OSFC) tissue from the patients who were treated in Liyuan Hospital were collected as OSFC group; 30 cases of oral submucous fibrotic(OSF) tissue from the patients who were treated in Liyuan Hospital were collected as OSF group; at the same time, 30 cases of normal oral mucosa, without betel nut and tobacco and alcohol addiction, were taken as normal group. Western blot was used to detect the relative expression of PTPRZ1 and hTERT; enzyme-linked immunosorbent assay was used to detect the expression of COX-2; immunohistochemistry was used to detect the samples. The correlation among PTPRZ1, COX-2 and hTERT in the oral submucous fibrotic canceration was analyzed. Results: The relative expression of PTPRZ1, COX-2 and hTERT in OSF group was higher than that in normal group (P < 0.05); the relative expression of PTPRZ1, COX-2 and hTERT in OSFC group was higher than that in normal group and OSF group (P < 0.05). The relative expression of PTPRZ1, COX-2 and hTERT in poorly differentiated oral submucous fibrogenic carcinoma was higher than that in well differentiated and moderately differentiated oral submucous fibrogenic carcinoma (P < 0.05). The relative expression of PTPRZ1, COX-2 and hTERT in stage Ⅳ oral submucous fibrogenic carcinoma was higher than that in stage Ⅱ and Ⅲ oral submucous fibrogenic carcinoma (P < 0.05). The relative expression of PTPRZ1, COX-2 and hTERT was positively correlated in oral submucous fibrocarcinogenesis. Conclusion: PTPRZ1, COX-2, and hTERT were abnormally high expressed in the oral submucous fibrocarcinogenesis; PTPRZ1, COX-2, and hTERT were high expressed in the oral submucous fibrocarcinogenesis with low differentiation and high pathological stage, and PTPRZ1, COX-2, hTERT were positively correlated in the oral submucous fibrocarcinogenesis.
