摘要:诱导多能干细胞(IPSCs)是通过强制表达多能因子重编程为多潜能的体细胞。这种细胞具有与胚胎干细胞(ESCs)相同的多潜能性,并被认为是有伦理争议的ESCs的替代细胞。然而,使用基因组整合逆转录病毒或慢病毒将细胞重编程成IPSCs的传统方法,通过遗传修饰来重编程IPSCs增加了恶性转化的可能性,并成为其在临床环境中应用的障碍。因此,需要一种替代方法在不使用基因编辑的情况下重编程细胞,并且是通过更安全的方式来传递转录因子以诱导靶细胞上的多能性。许多研究已经证明蛋白转导技术可以通过传递重编程蛋白从人成纤维细胞和小鼠胚胎成纤维细胞产生IPSCs。本文就蛋白转导技术在IPSCs重编程中的应用进展作一综述。 |

ABSTRACT: Induced pluripotent stem cells (iPSCs) are pluripotent somatic cells reprogrammed by forced expression of pluripotency factors. These cells which are regarded as an alternative to the ethical arguments of ESCs have the same pluripotent potential as embryon- ic stem cells (ESCs).However, The use of genome-integrating retroviruses or lentiviruses to reprogram cells into IPSCs as a traditional method of reprogramming of IPSCs by genetic modification may increase the possibility of malignant transformation and has been a limi- tation in clinical applications. Hence, there is a need for an alternative way to reprogram the cells without using gene editing and to deliver transcription factors in a more secure way to induce pluripotency on target cells. Numbers studies have shown that protein transduction technology can generate IPSCs from human fibroblasts and mouse embryonic fibroblasts by delivery of reprogramming proteins. In this paper, the application of recombinant protein-induced iPSCs in protein transduction technology was reviewed. |
