摘要 目的:探讨二甲双胍联合卡铂对三阴性乳腺癌细胞MDA-MB-231增殖的影响。方法:体外培养三阴性乳腺癌细胞MDA-MB-231,分别给予不同浓度(0、2.5、5、10 mmol/L) 二甲双胍和20 μmol/L卡铂处理后,采用MTT实验、平板克隆实验检测二甲双胍联合卡铂对MDA-MB-231细胞增殖的影响。结果:MTT实验结果显示:二甲双胍抑制MDA-MB-231细胞的增殖,5、10 mM组细胞OD490值均显著低于对照组(P <0.05),且10 mM组细胞OD490值明显低于其他各组(P<0.05);与单药相比,二甲双胍和卡铂联用组细胞生长抑制率显著升高,差异具有统计学意义(P<0.05)。平板克隆实验结果显示:与单药相比,二甲双胍和卡铂联用组细胞的克隆形成抑制率率显著升高,差异具有统计学意义(P<0.05)。结论:二甲双胍联合卡铂能够有效的抑制三阴性乳腺癌细胞MDA-MB-231的增殖。 |

ABSTRACT Objective: To explore the effect of metformin combined with carboplatin on the proliferation of triple negative breast cancer cells MDA-MB-231. Methods: MDA-MB-231 cell was cultured in vitro and treated with different concentrations (0, 2.5, 5, 10 mmol/L) of metformin and 20 μmol/L carboplatin, respectively. MTT and clone formation assays were used to detect the effect of metformin combined with carboplatin on the proliferation of MDA-MB-231 cells. Results: MTT assay showed that metformin inhibited the proliferation of MDA-MB-231 cells. The cells OD490 values in 5 or 10mM group were significantly lower than that of the control group (P <0.05), and the OD490 values in 10 mM group were significantly lower than those in the other groups (P <0.05). Compared with monotherapy, the cell growth inhibition rate of metformin combined with carboplatin was significantly increased(P<0.05). The results of plate clone formation assay showed that compared with monotherapy, the cell clone formation inhibition rate of metformin combined with carboplatin was significantly increased(P<0.05). Conclusion: Metformin combined with carboplatin could effectively reduce the proliferation of triple negative breast cancer cell line MDA-MB-231. |
