摘要 目的:研究鞣花酸对高脂饮食引起的大鼠肥胖预防的影响。方法:将40只SD 雄性大鼠按体重随机分为鞣花酸高剂量组(高脂饮食 + 30 mg/kg/d 鞣花酸,EAH),鞣花酸低剂量组(高脂饮食 + 10 mg/kg/d 鞣花酸,EAL),阳性对照组(高脂饮食 + 37.8 mg/kg/d 奥利司他,O),模型对照组(高脂饮食 + 10 mL/kg/d 0.5 % CMC钠溶液,HFD)和空白对照组(普通饮食 + 10 mL/kg/d 0.5 % CMC钠溶液,ND),连续给药10周,测量体重、进食量、体脂、肝重、肾重,计算脏器系数和脂肪系数,测定血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白胆固醇(LDL - C)和高密度脂蛋白胆固醇(HDL - C);HE染色观察脂肪细胞和肝脏组织细胞形态。结果:干预10周后,与模型对照组比较,鞣花酸高、低剂量组大鼠体重和进食量均显著下降(P <0.05),脂肪系数显著降低(P <0.001);鞣花酸高、低剂量组血清HDL-C增加(P <0.01),鞣花酸高剂量组血清TC降低 ( P <0.05);HE染色观察发现鞣花酸高、低剂量组脂肪细胞直径显著变小,肝组织脂滴空泡明显减少,且呈剂量依赖性。结论:鞣花酸可能通过抑制高脂饮食大鼠的摄食量从而抑制其体重的增加。 |

ABSTRACT Objective: To investigate the effect of Ellagic Acid on obesity prevention. Methods: Forty male SD rats were randomly divided into 5 groups ( n = 8 ) as follows: rats fed with HF diet with high dosage of Ellagic Acid( 30 mg/kg/d , EAH group), rats fed with HF diet with low dosage of Ellagic Acid ( 10 mg/kg/d, EAL group), rats fed with HF diet with Orlistat Capsules( 37.8 mg/kg/d , O group), rats fed with HF diet with placebo pellet(0.5 % CMC Na solution, 10 mL/kg/d , HFD group), and rats fed with normal diet with placebo pellet(0.5 % CMC Na solution, 10 mL/kg/d , ND group) for 10 weeks. Body weight, food intake, the index of body adipose tissue, liver and kidney were measured. The serum lipid level of TG, TC, HDL-C and LDL-C were analyzed. Adipocyte and liver cells were examined under a light microscope after HE staining. Results: Ellagic Acid high and low dosage could significantly decrease body weight, food intake and body adipose tissue index compared with HFD group (P < 0.05). All dosage of Ellagic Acid could significantly increase serum HDL-C(P < 0.01 or P < 0.05), and the high dose Ellagic Acid could significantly decrease serum TC(P < 0.05). The size of lipocytes of adipose tissue and the number of lipid droplet of liver of the high and low dosage of Ellagic Acid became smaller and littler. Conclusion: Ellagic acid could significantly inhibit body weight by inhibiting food intake in rats fed with high fat diet. |
