目的:研究Cytohesins介导EGFR 通路在结直肠癌细胞增殖中的作用。方法:选择人结直肠癌细胞系HT-29 作为研究细胞,
采用免疫荧光检测结直肠癌细胞系HT29 中EGFR 表达情况,SecinH3 阻断结直肠癌细胞系HT29 细胞内cytohensins 观察细胞
内EGFR 通路激活情况,MTT 法检测细胞增殖情况。结果:EGF+SecinH3 组细胞内p-EGFR 水平显著低于EGF组(P<0.05)。第
3、5 天,各组细胞增殖情况存在显著差异(F=38.072,P<0.05),其中EGF 组显著高于SecinH3 阻断组(P<0.05)和对照组(P<0.05),
对照组吸光度值显著高于SecinH3 阻断组(P<0.05)。结论:EGF 与结直肠癌细胞的增殖密切相关,通过阻断Cytohesins 介导
EGFR 通路能够显著抑制结直肠癌细胞的增殖。 |

Objective:To study the effect of cytohesins mediated EGFR pathway on the proliferation of colorectal cancer cells.Methods:Human colorectal cancer cell line HT-29 was set as the study cells. The expression of EGFR of colorectal cancer cell line HT29
was evaluated by immunofluorescence detection, and the activation of EGFR pathway was evaluated after SecinH3 blocked, and the cell
proliferation was detected by MTT method.Results:The p-EGFR level in EGF + SecinH3 group was significantly lower than that of EGF
group (P<0.05). After 2, 3 days of treatment, the difference of cell proliferation were statistically significant among three groups (F=38.
072, P<0.05), and it was significantly higher in the EGF group than that of SecinH3 group and control group (P<0.05), and the
absorbance value was significantly higher in control group than that of SecinH3 blocked group (P<0.05).Conclusion:EGF is closely
related with the colorectal cancer cell proliferation, which can significantly inhibit the proliferation of colorectal cancer cells by blocking
cytohesins mediated EGFR pathway. |
