Objective:To investigate the transformation of microglia (MG) to dendritic cells (DC) in the process of cerebral
ischemia reperfusion injury (CIRI).Methods:CIRI mouse model was established by thread embolism method. Immunofluorescence
staining was used to detect the transformation of MG into DC. Flow cytometry was used to detect the activation of MG, transformation of
MG into DC as well as the MHC-II expression on DC transformed from the MG.Results:MG (CD11b+) expression DC (CD11c+)
surface markers during CIRI. Compared with Sham group, activated MG (CD11b+CD45med) increased significantly(P1d<0.01, 2d, 4d,
6d group P<0.001), the number of activated MG transformed into DC (CD11b+CD45medCD11c+) was significantly increased (P<0.
001), which reached its peak at day 4, and was significantly increased compared with 1d, 2d, 6d group (P<0.001, P<0.01, P<0.05) in
brain tissue of CIRI mice. The expression of MHC-II was significantly increased (P<0.001) in the DC transformed from MG.
CD11b+CD45medCD11c+MHC-II+ cell number was significantly increased in 4d group compared with 1 d, 2 d, 6 d group (P<0.001,
P<0.001, P<0.05).Conclusion:MG can transforminto DC during CIRI, this kind of DC has the function of antigen presentation. |
