Objective:To study the effects of miR-155 on the cell cycle of prostate cancer and the underlying mechanisms.Methods:The cell cycles of DU145 and PC-3 cells were evaluated after anti-miR-155 transfection by FACS. p53 and p21 expression were evaluated
by western blot and RT-PCR.Results:The cell cycle of DU145 and PC-3 cells were down regulated obviously after anti-miR-155
transfection(P<0.05), and the protein and mRNA expression of p53 and p21 were obviously increased (P<0.01). In addition, the protein
and mRNA expression of CDK2 and cyclin E were also decreased obviously (P<0.01).Conclusion:Inhibition of miR-155 could modulate
the cell cycles of DU145 and PC-3 cells in vitro by p53/p21 pathways and by down regulating the expression of CDK2 and cyclin E. |
