目的:探讨miR-203 在肝细胞肝癌(Hepatocellular carcinoma,HCC)患者血浆中的表达及其对肝癌细胞增殖和侵袭能力的影
响。方法:选择2013 年5 月-2015 年7 月在我院确诊为肝细胞肝癌的患者57 例作为研究对象,另选取同期在我院接受健康体检
的志愿者49 例作为对照组。采用实时荧光定量RT-PCR 法检测两组血浆中miR-203的表达水平;将过表达的miR-203 转染至人
肝癌细胞系Hep 3B 和JHH-1;采用CCK-8 法检测miR-203 对肝癌细胞增殖能力的影响;采用transwell法检测miR-203 对肝癌
细胞迁移及侵袭能力的影响。结果:miR-203 在肝癌患者血浆中的表达水平显著低于对照组,差异具有统计学意义(P<0.01);与未
转染的肝癌细胞比较,Hep 3B 和JHH-1 细胞转染miR-203mimic 后,肿瘤细胞的增殖、迁移及侵袭能力受到明显抑制,差异具有
统计学意义(P<0.05)。结论:miR-203 在肝细胞肝癌中呈低表达,而过表达miR-203 能够抑制肿瘤细胞的增殖及侵袭能力,提示
miR-203 可以作为一种新的肿瘤抑制性microRNA。 |

Objective:To investigate the expression of miR-203 in HCC, and the effects on the proliferation and migration of tumor
cells.Methods:57 cases with HCC who were diagnosed in our hospital from May 2013 to July 2015 were selected as the research
objects, and another 40 healthy volunteers were chosen to be the healthy control group. The RT-PCR was used to detect the expressions
of miR-203 in the two groups. Then the over-expressed miR-203 was transfected into the human hepatocellular carcinoma cell line hep
3b and JHH-1. The effect of miR-203 on the proliferation of hepatoma cells was detected by CCK-8 assay, and the effect of miR-203 on
the migration and invasion was detected by Transwell assay.Results:The expression of miR-203 in HCC was significantly lower than
that of the control group, and the difference was statistically significant (P<0.01). Overexpression of miR-203 in HCC cells Hep 3B and
JHH-1 could inhibite the cells' proliferation, migration and invasion, and the difference was statistically significant (P<0.01).Conclusion:The expressionsof miR-203 in HCC was lower and the overexpression of miR-203 in HCC cells can inhibite the cells proliferation, migration
and invasion, which might be an tumor suppressive microRNA. |
