      目的:基于芯片数据库分析的基础上,探讨miR-32 在非小细胞肺癌患者中的表达水平。方法:在GEO和ArrayExpression 数 据库中搜索关键词"microRNA 和肺癌" 检索出两个样本量最大的芯片数据,分别是GEO 数据库中编码为GSE61741 和 ArrayExpression 数据库中编码为E-TABM-22 的芯片数据,对两组数据进行分析找出有差异的microRNA。另外收集 2010.02-2012.09 期间在吉林大学附属中日联谊医院胸外科进行肺癌手术切除的32 名患者的肺癌组织及配对的癌旁正常肺组织 标本,检测标本中miR-32 的表达水平。结果:综合分析GSE61741 和E-TABM-22 的芯片数据发现有共同差异的microRNA共有 8 个,其中上调的有2 个,分别是hsa-miR-192 与hsa-miR-197,下调的有6 个,分别是hsa-miR-126、hsa-miR-199b-3p、 hsa-miR-219-1-3p、hsa-miR-26a、hsa-miR-32 与hsa-miR-9。为了进一步探讨miR-32 在癌症中的作用,我们对GSE61741 中其他癌 症患者及健康者血浆中miR-32 的芯片数据进行分析,发现miR-32 在结肠癌、神经胶质瘤、肾癌、前列腺癌、Wilm's 瘤中的表达水 平均显著低于健康者(P<0.01)。对32 例及E-TABM-22 芯片数据中65 例NSCLC 患者癌及配对的癌旁正常肺组织中miR-32 的 表达水平分析发现癌组织中miR-32 的表达水平显著下调(P<0.001)。结论:miR-32 在非小细胞肺癌组织中低表达,提示miR-32 可能为新的NSCLC 诊断标记分子。

      Objective:To explore the expression of miR-32 in NSCLC(non-small cell lung cancer)patients based on the analysis of gene chip in database.Methods:Two microRNA data of gene chip with largest samples were analyzed in our research through searching the key words "microRNA and lung cancer" from the GEO and Array Expression databases. They were GSE61741 and E-TABM-22 respectively. And the differential gene miR-32 was selected and analyzed, and miR-32 expression in lung cancer tissues and matched adjacent normal lung tissues from 32 NSCLC patients who received surgical resection was detected.Results:Comparing the datasets of GSE61741andE-TABM-22, 8 differential expressions of microRNAs were found to have the same level. Among them, 2 miRNAs (hsa-miR-192 and hsa-miR-197) were increased and 6 miRNAs (hsa-miR-126, hsa-miR-199b-3p, hsa-miR-219-1-3p, hsa-miR-26a, hsa-miR-32 and hsa-miR-9) were decreased. The level of miR-32 was significantly lower in colon cancer, glioma, renal cancer, prostate cancer and Wilm's tumor than that in normal controls (P<0.01).The level of miR-32 in cancer tissues was significantly down-regulated than that of matched adjacent normal tissues in NSCLC patients with statistical significance (P<0.001).Conclusion:The level of miR-32 expression is down-regulated in NSCLC cancer tissues, suggesting that miR-32 may be a new diagnostic marker for this disease.
