FHL2 是仅有四个半LIM结构域(FHL)蛋白家族的成员,目前在FHL 蛋白家族中研究最为广泛。FHL2 作为重要的衔接蛋白
和支架蛋白,主要通过LIM结构域介导蛋白分子间的相互作用以实现其生物学功能。基因在转录水平受多种肿瘤相关基因
的调控,如p53,血清应答因子等。FHL2 与恶性肿瘤的关系是近年来的研究热点,目前认为FHL2 能够作为癌蛋白及抑癌蛋白通
过不同机制广泛影响乳腺癌、胃肠道肿瘤、肝癌、前列腺癌等肿瘤的发生发展,并且在不同肿瘤中的表达具有组织特异性。本文就
FHL2 的结构特点、功能、转录调控及与肿瘤的关系几个方面展开综述,从而明确FHL2 在不同肿瘤中所发挥的作用及其分子生物
学机制将会为治疗相关肿瘤提供新的干预靶点。 |

The FHL2 protein belongs to the FHL protein (Four-and-a-half LIM-domain protein) family, and is the most
investigated member within the FHL family. FHL2 proteins participate in various biological functions as adaptor molecules and
scaffolding proteins, mainly through the LIM domains, which can regulate protein-to-protein interactions. gene is regulated by
various tumorigenesis-related genes, for example p53 and serum response factor (SRF). The relationship between FHL2 and malignant
cancers has become a hot topic recent years. Up to now, FHL2 is proved to influence the growth and development of wide types of
cancers as oncoprotein or tumor suppressor in breast cancer, gastrointestinal (GI) cancer, liver cancer, prostate cancer and other cancers
through different mechanisms, and the FHL2 protein is expressed in a tissue-specific manner in different cancers. In this review, the
structure, functions, transcription of FHL2 and its relationship with related cancers will be discussed in order to understand the role and
the underlying molecular mechanisms of FHL2 in different cancers and provide a new intervention target for treating relative cancers. |
