目的：通过对月经周期紊乱患者的细胞周期相关基因的表达水平进行分析，得出温阳化瘀法在此病上的治疗效果。方法：选取我院妇科收治的月经周期紊乱患者120 例，参照随机原则共分为2 组，其中西医治疗组59 例，给予醋酸甲羟孕酮片和克罗米芬口服；实验组61 例，在西医治疗基础上给予温阳化瘀中药治疗，每日1 剂；另外随机选取同期体检健康的60 名女性作为正常对照组，在治疗结束后，应用免疫组化方法对全部受试者进行p53、p21以及MDM2 蛋白表达检测，同时应用统计学软件对相关结果进行分析。结果：①p53 和p21蛋白的阳性表达主要在细胞核，MDM2蛋白阳性表达定位在细胞核和（或）胞质；② 正常子宫内膜组织中p53 蛋白阳性表达率为（36.67 %），p21 表达率为（33.33 %），显著高于月经周期紊乱患者，中药治疗组的p53 蛋白阳性表达率为（19.67 %），p21 达率为（18.03 %），显著高于西医治疗组，P<0.05，差异有统计学意义；③正常子宫内膜组织中MDM2 蛋白阳性表达率为（1.67 %），显著低于月经周期紊乱患者，中药治疗组的MDM2 蛋白阳性表达率为（32.77 %），显著低于西医治疗组 (50.85 %)，P<0.05，差异有统计学意义。结论：与健康妇女相比，月经周期紊乱患者的p53 和p21 蛋白阳性表达率显著降低，MDM2 蛋白阳性率显著升高，应用温阳化瘀法能够显著改善月经周期紊乱患者细胞周期相关基因的表达水平。

Objective:To analyze the expression level of the cell cycle related gene in patients with menstrual cycle disorder, and investigate the curative effects of Wenyang Huayu method in treatment of this disease.Methods:120 patients with menstrual cycle disorder were selected in our hospital, with reference to the principle of random they were divided into 2 groups. 59 cases in Western medicine treatment groups, were given Medroxyprogesterone Acetate Tablets and clomiphene citrate oral; and 61 cases in the experimental group, based on the western medicine, were given the traditional Chinese medicine treatment of Wenyang Huayu method, daily 1 agent. In addition, 60 healthy female during the same period were randomly selected as the control group. At the end of treatment, immunohistochemical method was used to detect the expression of p21, MDM2, P53, at the same time the related results were analyzed using statistical software.Results:① The positive expression of p21 and P53 was mainly in the nucleus, and the positive expression of MDM2 protein was localized in the nucleus and (or) cytoplasm; ②in the normal endometrial tissue, the positive expression rate of P53 protein was 36.67 %, and the p21 was 33.33 %, significantly higher than that in patients with menstrual cycle disorder, and P53 protein positive expression of experimental group was 19.67 %, p21 was 18.03 %, significantly higher than that of Western medicine therapy group, P<0.05, the difference was statistically significant; ③ in normal endometrial tissues, the positive rate of MDM2 protein was 1.67 %, significantly lower than that of menstruation disorder patients, and the positive expression of MDM2 protein of the TCMexperimental group (32.77 %) was significantly lower than that of Western medicine therapy group (50.85 %), P<0.05, the difference was statistically significant.Conclusion:Compared with healthy women, the expression of p53 and p21 protein was decreased in patients with menstrual cycle disorder, and MDM2 protein expression was significantly increased. The treatment of Wenyang Huayu method can significantly improve the disorder of cell cycle related gene in the patients with menstrual cycle disorder.
