法:根据人GDF15 氨基酸序列特征,设计合成了8 条能够免疫产生GDF15特异性抗体的抗原多肽,与VLP载体偶联后,免疫雌
性BALB/c 小鼠,利用杂交瘤技术制备鼠源抗人GDF15 的mAb,用间接ELISA 检测mAb 腹水效价。结果:获得针对7 个抗原多
肽的12 株稳定分泌抗人GDF15的杂交瘤细胞系,腹水mAb 效价可达1× 104~1× 109。结论:获得了针对不同抗原多肽的抗人
GDF15的特异性mAb,为进一步研发以GDF15 为靶点的单克隆抗体抗肿瘤药物奠定了基础。 |

Objective:To establish the specific hybridoma cell lines that secreted stably monoclonal antibody (mAb) against
growth differentiation factor-15 (GDF15) from human.Methods:Eight antigen peptides which can produce GDF15 specific antibodies
were designed and coupled with VLP carrier. Then female BALB/c mice were immunized with coupled VLP carrier and mAb against
GDF15 was prepared with hybridoma cell fusion technique. Indirect enzyme-linked immunosorben assary (ELISA) was used to evaluate
the ascites titers of hybridoma cell lines.Results:Twelve hybridoma cell lines which can stable produce anti-GDF15 mAb against seven
antigen peptides were obtained. The titers of mAb in ascites were ranged from 1× 104 to 1× 109.Conclusion:The specific mAb against
different antigen peptides of GDF15 were produced, which will be significant for monoclonal antibody research of GDF15-targeted
drugs. |
