receptor,LOX-1)表达。方法:不同浓度的Ox-LDL和培养的人肾小球系膜细胞共孵育,应用Real-time PCR和Western Blot 方法检
测Ox-LDL对人肾小球系膜细胞LOX-1 表达的影响。结果:Ox-LDL剂量和时间依赖性促进人肾小球系膜细胞LOX-1mRNA和
蛋白表达。Ox-LDL 40 ug/mL刺激细胞0-24 小时,于12 小时达峰值。Ox-LDL 10、20、40、60 ug/mL 分别作用于细胞12 小时,40
ug/mL组达到峰值,为基础值的3.73 倍。Ox-LDL 40 ug/mL刺激细胞0-24 小时,LOX-1 蛋白24 小时达高峰,Ox-LDL 10、20、40、
60 ug/mL分别作用细胞24 小时,40 ug/mL组细胞LOX-1 蛋白达到峰值,为基础值的1.81 倍。结论:Ox-LDL 在一定浓度范围内

Objective:To observe whether the expression of lectin-like oxidized LDL receptor can be induced by Ox-LDL on
human mesangial cell (HMC).Methods:When cultured HMC stimulated by different doses of Ox-LDL, the levels of LOX-1 on HMC
induced by Ox-LDL were examined using Real-time PCR and Western Blot.Results:Ox-LDL could induce LOX-1 mRNA and protein
expression in a dose-dependent manner (10-40 ug/mL). The peak level of LOX-1 mRNA reached after 12 h of stimulation and was as
high as 3.73-fold as control. The up-regulation effect on LOX-1 protein reached peak after 24h and was as high as 1.81-fold as control
treated with 40 ug/ml of Ox-LDL.Conclusion:Ox-LDL can induce LOX-1 expression in a dose and time dependent manner. |
