Objective:To investigate the effects of different concentrations of sevoflurane and isoflurane on proliferation,
apoptosis and chemosensitivity to cisplatin of MG63 cells.Methods:MG63 cells were inoculated in culture plate. The proliferation
inhibition rates, colony formation rates and apoptosis percentages of MG63 cells were measured by methyl thiazolyl tetrazolium(MTT)
assay, colony formation assay and flow cytometer respectively after exposure to different concentrations of sevoflurane and isoflurane.Results:Sevoflurane and isoflurane inhibited to some extent, the proliferation of MG63 cells. Compared to that of other concentrations
(P<0.05), the inhibition effect of 5.1% Sevoflurane and 2.6% Isoflurane was more obvious (P<0.05). Sevoflurane and isoflurane
inhibited the colony formation of MG63 cells (P<0.05). Different concentrations of sevoflurane and isoflurane had no effect on
apoptosis of MG63 cells(P>0.05). 2.5%Sevoflurane and 2.0%Isoflurane decreased the apoptosis percentages induced by cisplatin on
MG63 cells.Conclusion:Different concentrations of sevoflurane and isoflurane can inhibit the proliferation and colony formation of
MG63 cells, and it is more obvious to the proliferation inhibition rates of MG63 cells by high concentrations of Sevoflurane and
Isoflurane. Sevoflrane and isoflurane don't promote the apoptosis of MG63 cells, but can reduce its chemosensitivity to cisplatin. |
