      目的:研究不同强度脉冲电磁场干预对成肌细胞增殖的影响。方法:10Hz脉冲低频电磁场刺激经复苏后培养贴壁良好的C2C12成 肌细胞,根据不同磁场强度和作用时间将其分为A、B、C 组,无磁场干预的为对照组。采用RT-qPCR检测不同磁场强度下成肌细 胞标记基因Myf5、MyoD及Pax7 的mRNA 的表达。结果:经RT-qPCR 检测三种基因的表达情况,Myf5 mRNA 在1.5 mT 磁场强 度下照射第五天表达最高;0.5 mT 磁场强度下Myf5 mRNA 的表达与对照组相比无统计学意义(P>0.05);1.0 mT 磁场强度下 Myf5 mRNA 表达与对照组比较差异具有统计学意义(P<0.05);1.5 mT 磁场强度下Myf5 mRNA表达与对照组比较差异具有统计 学意义(P<0.05)。对照组MyoD mRNA的表达要比磁场作用下表达要高。三个磁场强度下MyoD mRNA 表达与对照组相比均无 统计学意义(P>0.05)。0.5 mT、1.0 mT 磁场强度下Pax7 mRNA 的表达要比对照组要高,与对照组相比具有统计学意义(P<0.05); 1.5 mT磁场强度下Pax7 mRNA表达与对照组相比无统计学意义(P>0.05)。结论:1.5 mT 脉冲电磁场强度下作用5 天对体外培 养的成肌细胞Myf5 mRNA 标记基因增殖促进作用最强。

      Objective:To study the effects of different intensity pulsed electromagnetic field interference on myoblast proliferation.Methods:Four groups were divided in the experimen: control group without intervention field; group A, in which the magnetic field strength was 0.5mT experimental, group B 1.0mT experimental, group C 1.5 mT experimental. Expression of marker gene Myf5, MyoD and Pax7 of mRNA was detected using RT-qPCR under different magnetic field strength myoblast.Results:By RT-qPCR detection of the expression of three genes, the expression of Myf5 mRNA was the highest on the fifth day of the irradiation field strength 1.5mT; the expression of Myf5 mRA in the group magnetic field strength was 0.5 mT, which, compared with in the control group, was not statistically significant (P>0.05); the expression of Myf5 mRNA in the group magnetic field strength was 1.0 mT, compared with in the control group was statistically significant (P<0.05); the expression of Myf5 mRNA in the group magnetic field strength is 1.5 mT, compared with in the control group, it was statistically significant (P<0.05). The expression of MyoD mRNA in the control group was higher than the magnetic field. Under the three magnetic fields, strength MyoD mRNA expression, compared with in the control group, was not statistically significant (P>0.05). The expression of Pax7 mRNA in the group magnetic field strength was 0.5 mT and 1.0 mT, compared with that in the control group, it was statistically significant (P< 0.05); the expression of Pax7 mRNA in the group magnetic field strength was 1.5mT, compared with that in the control group, it was not statistically significant (P>0.05).Conclusion:1.0 mT and 1.5 mT pulsed electromagnetic fields on cultured myoblasts are with proliferation, but the growth promoting of cultured myoblasts marker gene Myf5 mRNA is the strongest at 10 Hz, 1.5 mT pulsed electromagnetic field strength.
