      目的: 观察下丘脑腹内 侧核( VMH)损毁对大鼠脂肪组织 nesfatin-1/NUCB2 表达的影响及其机制。 方法: 电损毁 VMH, 观察 大鼠体重和脂肪组织变化,采用 Western blot 检测脂肪组织中 nesfatin-1 /NUCB2 表达改变。 腹腔注射 6- 羟多 巴胺( 50 mg/kg)以阻 断交感神经;持续外周注射卡巴胆碱( 1 80 滋g/kg)用 以模拟 VMH 损毁,观察其对大鼠皮下脂肪 nesfatin-1 /NUCB2 表达的影响。 结 果: 与对照组和假手术组比较, VMH 损毁后大鼠体重明显增加( P<0.05), 皮下脂肪( P<0.05)和肠系膜脂肪( P<0.05)也显著增多 ; Western blot 分析结果显示, nesfatin-1/NUCB2 在胰腺和肝脏中表达较多 , 但皮下、肠系膜脂肪和肩胛间棕色脂肪组织( iBAT)中表 达较少 , 骨骼肌( 腓肠肌)中 鲜有表达; 与对照组和假手术组比较, VMH 损毁组大鼠 nesfatin-1/NUCB2 在肝脏、 胰腺、 骨骼肌 和 iBAT 中表达无显著差异( P>0.05),皮下脂肪( P<0.05)和肠系膜脂肪( P<0.05)nesfatin-1 /NUCB2 表达显著增多 与对照组相比, 6- 羟多 巴胺组 nesfatin-1/NUCB2 表达显著升高( t=3.43, P<0.05),而卡巴胆碱组 nesfatin-1/NUCB2 表达无显著差异( t=0.37, P=0.72)。 结论: VMH 损毁后大鼠脂肪组织 nesfatin-1 /NUCB2 表达改变可能通过抑制交感神经活动介导。

      Objective:To observe the effects of ventromedial hypothalamus nuclear lesion on nesfatin-1 /NUCB2 expression of white adipose tissue in rats, and to explore its underlying mechanism.Methods:The hypothalamic ventromedial nucleus (VMH) of rats was damaged by electrical stimulation. The changes of weight and adipose tissues of rats were observed. And the expression of nesfatin-1 /NUCB2 in adipose tissues was detected by Western blot analysis. Intraperitoneal injection of 6-hydroxy dopamine (50 mg/kg) was used to block the sympathetic nerve; continuous peripheral injection of kappa choline (180 滋g/kg) was used to simulate VMH damaged to observe the nesfatin-1/NUCB2 expression in subcutaneous fat of rats.Results:Compared with the control group and sham-operated group, the weight (P<0.05), subcutaneous fat (P<0.05) and mesenteric fat (P<0.05) of VMH-lesioned rats increased significantly. The results of Western blot analysis showed that nesfatin-1 /NUCB2 was mainly expressed in the pancreas and liver, less in subcutaneous, mesenteric fat and interscapular brown adipose tissues (iBAT), little in skeletal muscle (calf). Compared with the control group and sham-operated group, the expression of nesfatin-1/NUCB2 in liver, pancreas, skeletal muscles and iBAT in VMH-lesioned group had no significant differences, but in subcutaneous fat (P<0.05) and mesenteric fat (P<0.05) it significantly increased. Compared with that in the control group, the expression of nesfatin-1 /NUCB2 in 6-hydroxy dopamine group increased significantly (t=3.43, P<0.05), but was reduced in carbachol group (t=0.37, P=0.72).Conclusion:These results show that after VMH lesion, the increased expression of nesfatin-1 /NUCB2 in white adipose tissues may be mediated via inhibition of the sympathetic nerve activity.
