目的:研究甘油三酯代谢过程对紫外线照射后HaCaT细胞的影响及机制研究。方法:HaCaT 细胞分为4 组,分别为对照组、
紫外线照射组、紫外线照射并予甘油三酯组、紫外线照射并予甘油三酯合成酶抑制剂TOFA 组,查看MMP-1、COX-2 和IL-1beta在
mRNA 及蛋白水平上的变化,并查看其上游转录因子NF-κ B/Iκ B的表达情况。结果:1)甘油三酯可以抑制紫外线照射后细胞外
基质溶解蛋白MMP-1的表达(P<0.05)。2)甘油三酯可以抑制紫外线照射后炎症相关蛋白COX-2 和IL-1beta的表达(P<0.05)。3)甘
油三酯合成酶抑制剂TOFA 可以增强外线照射后细胞外基质溶解蛋白MMP-1以及炎症相关蛋白COX-2 和IL-1beta的表达(P<
0.05)。4)其调节过程可能通过NF-κB/IκB 信号传导通路实现(P<0.05)。结论:甘油三酯对紫外线的照射后的HaCaT 细胞有保护作
用,若抑制其代谢将减弱其保护作用。 |

Objective:To investigate the effects of Triglycerides (TG) metablism on the cultured HaCaT cells after ultraviolet
radiation and the possible mechanism.Methods:The HaCaT cells were divided into 4 groups, group A was control group, group B was
treated with ultraviolet radiation, group C was treated with TG before radiation and group D was treated with inhibitor of TG
synthase-TOFA before radiation. And the expression of mRNA and protein levels of MMP-1, COX-2 and IL-1beta were observed after the
treatment. The expression of upstreamtranscript factor NF-kB/IkB was also detected.Results:1) The expression of MMP-1 was inhibited
after the treatment of TG (P<0.05). 2) The expression of COX-2 and IL-1beta was inhibited after the treatment of TG (P<0.05). 3) TG
synthase inhibitor TOFA enhanced the expression of MMP-1, COX-2 and IL-1beta(P<0.05). 4) The process might be regulated by
NF-kB/IKB signaling pathway (P<0.05).Conclusion:TG has protective effect on HaCaT cells radiated by ultraviolet light which effect
can be decreased after inhibition of TG synthesis. |
