      摘要 目的: 探讨依达拉奉对犬自体肾移植缺血再灌注损伤的影响及机制。方法: 将 18 只体重匹配的健康杂种犬随机分为假手术 组(S 组)、 依达拉奉组(ED 组)、 生理盐水组(PS 组), 每组各 6 只。S 组仅进行肾手术切除; ED 组在阻断前在供体静脉内注入依达拉 奉 10 mg/kg, 然后使用 200 mL 加入依达拉奉 10 mg/kg 的 UW 液灌洗供体肾并在同样的保存液中保存供体肾 8 小时, 且再灌注 开始时立即在受体静脉内给予依达拉奉 10 mg/kg; PS 组同法使用相同体积的生理盐水。再灌注 4h 后检测 MDA、 MPO、 SOD、 iNOS、 eNOS 等活性; 术后 24h 检测血清肌酐(Cr)、 尿素氮(BUN)浓度; 光镜下观察肾组织病理学改变。结果: PS 组 MDA 显 著高于 S 组及 ED 组 (P<0.05), PS 组 MPO 含量亦低于 S 组及 ED 组 (P<0.05)。PS 组 SOD、 eNOS 含量显著低于于 S 组及 ED 组 (P<0.05), PS 组 iNOS 含量高于 S 组及 ED 组(P<0.05), ED 组的肾损伤明显减轻。结论: 依达拉奉能减轻肾移植缺血再灌注损伤, 其机制可能与减轻肾脂质过氧化反应有关。

      ABSTRACT Objective:To investigate the effect of edaravone on malonyldialdehyde (MDA), myeloperoxidase (MPO), renal tissue superoxide dismutase (SOD), inducible nitric oxide synthase (iNOS), endothelial nitric oxide synthase (eNOS) after canine autologous renal transplantation. Methods:18 weight matched healthy mongrel dogs were randomly divided into sham operation group (group S), edaravone group (group ED) and normal saline group (group PS), with 6 cases in each group. Group S: only kidney operation resection. Group ED: before clamping in the donor intravenous edaravone 10mg/kg, UW lavage and then use the 200 mL to join the edaravone 10mg/kg donor kidney and the preservation of donor kidney preservation solution for 8 hours in the same, and at the beginning of reperfusion immediately given edaravone 10 mg/kg in receptor intravenous. Group PS:with the same method using the same volume of saline. We detected the activities of MDA, MPO, SOD, iNOS and eNOS after reperfusion for 4H, and postoperative 24h detection of serum creatinine (Cr), blood urea nitrogen (BUN) concentration, while pathological changes of renal tissue in each group were observed under light microscope.Results: In PS group, MDA was significantly higher than those in S group and ED group (P<0. 05), iNOS of PS group were also lower than those inS group and ED group (P<0.05). In group PS, SOD, eNOS content was significantly lower than those in S group and ED group (P<0.05), iNOS of PS group was higher than those in S group and ED group (P<0.05), the pathological injury in ED group was significantly reduced.Conclusion: Edaravone can reduce ischemia reperfusion injury in renal transplantation, and its mechanism may be related to reduce renal lipid peroxidation.
