法:通过O/W 乳液法静电纺丝制备纳米纤维,并用扫描电镜对纳米纤维表面进行了表征。以GM-CSF 为模型蛋白,采用ELISA
双抗体夹心法考察纤维的体外释放行为;以BSA 为模型蛋白,用SEC-HPLC 比较纤维制备前后蛋白的聚集情况;以β- 半乳糖苷
酶为模型蛋白,用ONPG 法比较纤维制备前后酶的催化活性。结果:纤维表面平滑,直径均一,呈现互相连通的三维网状结构。纤
维在5 天内释放90%以上;纤维中回收的BSA 单体比例为66.53%;β- 半乳糖苷酶在纤维中的催化活性保持原活性的3.37%。结

Objective: To investigate the method of detecting the in vitro release and protein bioactivity of electrospun nanofibers
with different model proteins. Methods: Nanofibers were prepared byW/O emulsion electrospinning. ELISA method was used to investigate
the in vitro release of nanofibers with GM-CSF as model protein. SEC-HPLC was used to test the protein aggregates with BSA as
model protein. ONPG method was used to test protein bioactivity with β-galactosidase as model protein. Results: The nanofibers were
smooth and uniform. GM-CSF can be released more than 90% in 5 days; BSA monomer preserved 66.53% after recovering from
nanofibers; β-galactosidase bioactivity preserved 3.37% after recovering from nanofibers. Conclusion: The in vitro release and protein
bioactivity of electrospun nanofibers can be tested fast and accurate with different model proteins. |
