织,采用RT-PCR 检测两组ATF5、Bcl-2 mRNA 的表达情况,并分析其表达与肿瘤分化程度的相关性。结果:ATF5、Bcl-2 在结直肠
腺癌组织中的表达均高于癌旁组织(P<0.01);ATF5、Bcl-2 mRNA 表达高低与结直肠腺癌细胞分化程度相关,ATF5 mRNA 在低
分化组表达高于高分化组(P<0.05),Bcl-2 mRNA 在高分化组表达高于低分化组(P<0.05);ATF5、Bcl-2 mRNA 在结直肠腺癌组织
内表达呈负相关。结论:ATF5 在结直肠癌的发生、发展中具有一定作用,其作用机制可能与调节其下游基因Bcl-2 的表达有关。 |

Objective: To investigate the expression of Activating Transcription Factor 5 (ATF5) mRNA and B-cell lymphoma/
Leukemia-2 (Bcl-2) mRNA in colorectal adenocarcinoma. Methods: RT-PCR was used to examine the expression level of ATF5
and Bcl-2 mRNA and in 36 colorectal adenocarcinoma tissues and adjacent tissues. Then the relationship between ATF5 and Bcl-2 was
analyzed. Results: The mRNA expression level of ATF5 and Bcl-2 in colorectal adenocarcinoma tissues were higher than the adjacent
tissues (P<0.01), and in the adenocarcinoma tissues, the mRNA levels were related to the differentiation of the colorectal tumors. More
ATF5 mRNA was detected in poorly differentiated adenocarcinoma tissues than well/moderately differentiated adenocarcinoma tissues
while the Bcl-2 with a completely opposite result. A negative correlation between ATF5 and Bcl-2 was found in the colorectal adenocarcinoma
tissues. Conclusion: The abnormal expression of ATF5 may play an important role in the development and progression of colorectal
adenocarcinoma. ATF5 may play it's role as a inhibitor of apoptosis by regulating the expression of Bcl-2. |
