      目的:研究重组人粒细胞集落刺激因子(rhG-CSF)动员对供者CD4+ T 细胞表面分子淋巴细胞功能相关抗原-1(LFA-1)、细 胞间黏附分子-1(ICAM-1)、L- 选择素(LAM-1)和人整合素-4(VLA-4)的表达及其介导的CD4+ T 细胞功能的影响,探讨外周血 干细胞移植过程中CD4+ T 细胞免疫耐受机制。方法:使用三色荧光标记检测动员前及动员后第5 天供者外周血LFA-1、 ICAM-1、LAM-1 和VLA-4 的表达率,ELISA 方法检测动员前后CD4+ T 细胞分泌IFN-γ 和IL-4 能力,免疫磁性分选法分离纯化 CD4+ T 细胞,检测动员前后CD4+ T 细胞对基质细胞衍生因子-1α(SDF-1α)的迁移能力和对ICAM-1 的黏附能力。结果:动员前 后CD4+ T 细胞LFA-1(CD11a)和VLA-4(CD49d)表达率差异无统计学意义(P>0. 01),动员前后CD4+ T 细胞LAM-1(CD62L)和 ICAM-1(CD54)的表达率差异均有统计学意义,动员前显著高于动员后(P<0.01);动员前后CD4+ T 淋巴细胞向SDF-1α 的迁移率 差异无统计学意义(P>0. 01);动员后CD4+ T 细胞对ICAM-1 的黏附率降低(P<0.01);动员后IL-4 和IFN-γ 两个细胞因子在外周 血血清的浓度均降低(P<0.01)。结论:rhG-CSF 动员不影响CD4+ T 细胞LFA-1 和VLA-4 表达及CD4+ T 细胞迁移,但影响CD4+ T 细胞ICAM-1 和LAM-1 表达以及CD4+ T 细胞通过LFA-1 对ICAM-1 的黏附能力影响,并可能影响CD4+ T 细胞分泌细胞因子 IL-4 及IFN-γ 的功能。

      Objective: To investigate the mechanism of CD4+ T cells immunotolerance during allogeneic peripheral blood stem cell transplantation by studying the impact of recombinant human granulocyte colony stimulating factor (rhG-CSF) on expression of lymphocyte function antigen-1 (LFA-1), intercellular adhesion molecule (ICAM-1), L-Selectin (LAM-1) and very late antigen 4α (VLA-4) on CD4+ T cells, and functions of CD4+ T cells through these molecules during mobilization. Methods: The expression of LFA-1, ICAM-1, LAM-1 and VLA-4 of CD4+ T cells were determined by FACS before rhG-CSF mobilization and the fifth day of mobilization. The expression of IFN-γ and IL-4 by CD4+ T cells were examined with ELISA. At the same time, the migration and adhesion activity of CD4+T cells to SDF-1αand ICAM-1 were detected. Results: The expression of LFA-1 on CD4+ T lymphocytes were both 100%. The expression of VLA-4 didn't change markedly before and the fifth day of mobilization (P>0. 01). The expression of LAM-1 (CD62L) and ICAM-1 (CD54) on CD4+ T lymphocytes decreased during mobilization (P<0.01). SDF-1α induced CD4+ T lymphocytes migration for 4 hours didn't change markedly during mobilization (P>0.10). The adhesion activity of CD4+T lymphocytes to ICAM-1 decreased markedly during mobilization (P>0.10). Conclusion: The concentration of IL-4 and IFN-γ in peripheral blood serum decreased markedly during mobilization (P<0.01). In conclusions, rhG-CSF does not have effect on the expression of LFA-1 and VLA-4 on CD4+ T cell, and the migration activity of CD4+ T cell during mobilization, but can affect the expression of ICAM-1and LAM-1 on CD4+ T cell, and the adhesion activity of CD4+ T cell to ICAM-1 through LFA-1.
