目的:研究丁咯地尔和α- 硫辛酸(ALA)对糖尿病(DM)大鼠肾功能的保护作用。方法:雄性SD 大鼠用链脲佐菌素(60 mg/kg)
腹腔注射诱导DM。大鼠随机分为健康组(NC)、DM 组(DMC)、DM+ALA 组(ALA 组)、DM+ 丁咯地尔组(丁咯地尔组)。ALA 组和丁
咯地尔组每天给予ALA 100 mg/kg 灌胃和丁咯地尔(0.1 g/kg)腹腔注射,NC 组和DMC 组大鼠给予等量生理盐水每日灌胃。干预
4 周后,测观察治疗后大鼠血糖、尿素氮、血肌苷,内生肌苷清除率、24 小时尿蛋白排泄率,比色法检测血清抗氧化酶和丙二醛
(MDA)含量。结果:①丁咯地尔和α- 硫辛酸降低糖尿病大鼠的尿素氮、血肌苷、24h 尿白蛋白排泄率、丙二醛,增加超氧化物歧化
酶②两种药物联合用药具有协同作用。结论:丁咯地尔和ALA 可能通过抑制机体氧化应激水平,减低早期DM 大鼠肾脏损害。 |

Objective: To investigate the protective effect of Buflomedil andα-lipoid acid on renocortical of diabetic rats. Methods:
Experimental diabetes model was induced by intraperitoneal injection of streptozotocin (60 mg/kg)in male Sprague Dawley rats. The rats
were randomly divided into following 5 groups: normal control(NC),diabetes mellitus control(DMC),diabetics treated with Buflomedil
(0.1 g/kg) intraperitoneal injection,and diabetics treated with αlipoid acid (ALA,100 mg/kg/day). After 4 weeks treatment, blood glucose,
24 h urinary protein (24 h UP),serum creatinine,and blood urea nitrogen were detected as renal function indexes. And the malondialdehyde
(MDA) and antioxidant enzymes was evaluated with colorimetry method. Results: ①Buflomedil andα-lipoid acid decrease
blood urea nitrogen, serum creatinine, and 24 h urinary protein (24 h UP), serum creatinine, MDA and blood urea nitrogen; increase
Serum superoxide dismutase②two drugs have a synergistic effect in combination. Conclusion: Buflomedil and ALAmay could inhibiting
the oxidative stress in renal cortex of diabetic rats,so that reducing the early kidney injury. |
