      目的:研究姜黄素对肺纤维化大鼠肺成纤维细胞增殖、凋亡的影响,探讨凋亡诱导因子(AIF)在肺成纤维细胞凋亡中的作 用。方法:将体外培养的肺纤维化大鼠成纤维细胞,分别于不同浓度的姜黄素(5、10、20、40 μM)和caspase-3 抑制剂Z-DEVD-fmk (20 μM)孵育,观测细胞生长状态变化。MTT 检测成纤维细胞增殖抑制率;流式细胞仪检测细胞凋亡率;Western-Blot 测定凋亡诱 导因子(AIF)蛋白表达及核转位结果:流式细胞术检测细胞凋亡,5~40 μM 姜黄素处理12 h,其凋亡率呈浓度依赖,对照组相 比,差异显著;而抑制caspase-3 并不能完全阻止细胞凋亡。Western-Blot 结果显示,姜黄素处理组出现凋亡诱导因子(AIF)蛋白表 达与核转位,抑制caspase-3 活性后未检测出AIF 表达结论:姜黄素可抑制肺成纤维细胞增殖,其诱导大鼠肺成纤维细胞凋亡,可 能与线粒体释放AIF 有关。

      Objective: To the effect of curcumin on proliferation and apoptosis and the mechanism of apoptosis-inducing factor (AIF) on rat pulmonary fibroblasts. Methords: Fibroblasts derived from lung tissue of rat were cultured in vitro. Cultured cells were exposed to curcumin with different final concentration (5 μM,10 μM,20 μM,40 μM) and Z-DEVD-fmk with final concentration(20 μM). MTT assay was used to determine the inhibitory rate of curcumin on the proliferation of cells. Flow cytometry identified the apop- tosis. Western-blot was used to determine the expression and nuclear translocation of AIF protein. Results: Curcumin inhibited prolifera- tion of pulmonary fibroblasts in a dose-dependent manner (5 μM,10 μM,20 μM,40 μM), which was detected by Flow cytometry. But the proliferation inhibitory and the apoptosis induced by curcumin were not inhibited by Z-DEVD-fmk. In curcumin treated groups, AIF was observed associated with the nuclear translocation. Conclusion: Curcumin inhibited proliferation of pulmonary fibroblasts. The curcumin- induced apoptosis may be associated with the releasing of AIF from mitochondria.
