      目的:探讨青岛地区汉族人群阿司匹林抵抗与血小板膜糖蛋白GPⅢaPLA、Ⅰa807C/T 基因多态性之间的关系。方法:筛选 150 例动脉粥样硬化患者服用阿司匹林(100mg/d)至少14d 以上,根据血小板聚集功能测定将其分为阿司匹林抵抗(AR)组、阿司 匹林半抵抗(ASR)组,阿司匹林敏感(AS)组。用PCR-RFLP 法确定各组GPⅢaPLA、GPⅠa 807C/T 基因型。结果:仅于ASR 组检出 1 例PLA1/A2 基因型,其余均为PLA1/A1 基因型,未发现PLA2/A2 基因型,差异无统计学意义(P>0.005);GPⅠa807C/T 基因位点 AR 组、ASR 组的T 等位基因频率均显著高于AS 组,有统计学意义(P<0.005)。结论:GPⅢaPLA2 基因可能不是阿司匹林抵抗的遗 传危险因素。而GPⅠa 807C/T 基因位点的T 等位基因与阿司匹林抵抗的发生相关联,可能是阿司匹林抵抗遗传易感因素。

      Objective: To investigate whether two Gene Polymorphisms of Platelet Glycoprotein, GPⅢa PLA and GPⅠa807C/T, contribute to aspirin resistance in Han population of Qingdao. Methods: 150 patients with artherosclerosis on aspirin(100mg/d) for at least 14 days were enrolled. According to the Platelet agglutination test, they were divided to three groups: aspirin resistant(AR) group, aspirin semiresponder (ASR) group and aspirin sensitive (AS) group, Genotyping of the two polymorphisms was performed using a polymerase chain reaction-based restriction fragment-length polymorphism analysis. Results: Only one patient carried the PLA1/A2 allele. The others possessed PLA1/A1 allele. None PLA2/A2 observed. No relation was found between resistance status and PLA genotype. For the 807C/T genotype, the frequency of T allele in AR group and ASR group were higher than that in AS group. The difference was statistically Significant( P<0.005). Conclusion: The present results suggest that the PLA polymorphism of glycoprotein Ⅲa is not a risk factor for AR. But the 807T allele of the GPⅠa gene show higher relationship with aspirin resistance. It may be a genetic factor for susceptibility to AR.
