      目的 探讨主要组织相容性复合物I类相关蛋白A/B(MICA/B)在宫颈病变组织及宫颈细胞系中的表达及定位。方法 采用免疫组织化学SP法检测宫颈炎症组织、宫颈鳞状上皮内病变(squamous intraepithelial lesions,SIL)及宫颈鳞癌(cervical squamous cell carcinoma,CSCC)组织中MICA/B蛋白的表达情况。采用免疫荧光化学与激光共聚焦显微术结合的方法研究3种宫颈癌细胞系C33a(HPV-)、Siha(HPV16+)、 Hela(HPV18+)及正常宫颈上皮细胞系H8中MICA/B的表达和定位。结果 MICA/B蛋白主要表达定位于细胞浆,部分细胞核,在癌组织中阳性表达率 (83.3%、81.8%) 明显高于宫颈炎症组织(39.3%、44.0%) , 差异具有统计学意义 (均有P<0.001) ; MICA蛋白在SIL组织的表达率(81.8%)高于宫颈炎症组织的表达率(39.3%),差异具有统计学意义(P=0.002);与分化程度、临床分期、淋巴结转移等临床病理参数之间比较无统计学意义(P>0.05)。结论 MICA蛋白随着宫颈组织病变的加重表达逐渐增高,提示MICA蛋白对宫颈病变的发生有重要意义。MICB蛋白在宫颈癌组织的表达高于宫颈炎症组织组织,提示MICB蛋白可能与宫颈癌的发生有关。

      Objective:To investigate the expression and localization of major histocompatibility complex (MHC) class I chain-related proteins A/B in cervical lesions tissues and cervical cancer cell lines. Methods:Immunochemistry SP method was used to detect the expression of MICA/B proteins in Cervical inflammation tissues 、SIL and CSCC. The localization of MICA/B in three cervical cancer cell lines C33a(HPV-)、Siha(HPV16+)、Hela(HPV18+)and normal cervical epithelial cell line H8 were detected by using immunofluorescence and Laser confocal microscopy. Results:MICA / B proteins are mainly expressed in the cytoplasm and some nucleu,The positive expression rates in cancer tissues (83.3%, 81.7%) were significantly higher than those in cervical inflammation tissues (39.4%, 44.0%),The difference was statistically significant (P <0.000, P <0.001).The expression rate of MICA protein in HSIL tissues (81.8%) is higher than that in normal cervical tissues (39.3%), and the difference is statistically significant (P = 0.002).There was no statistically significant difference with clinical pathological parameters such as the degree of differentiation, clinical stage, and lymph node metastasis (P> 0.05).Conclusion:The expression of MICA protein increases gradually with the aggravation of cervical lesions, suggesting that MICA protein may be involved in the occurrence of cervical lesions. The expression of MICB protein in cervical cancer tissues is higher than that in cervical inflammation tissues, suggesting that MICB protein may be related to the occurrence of cervical cancer.
