目的:考察金花茶二氯甲烷乙酸乙酯提取物对MMTV-PyMT转基因小鼠乳腺癌发生的实验预防作用。方法: MTT法检测二氯甲烷乙酸乙酯提取物对人乳腺癌细胞SKBR-3的体外增殖抑制作用;采用MMTV-PyMT转基因自发乳腺癌小鼠模型观察金花茶二氯甲烷乙酸乙酯对小鼠体内肿瘤发生的预防作用。结果:金花茶二氯甲烷乙酸乙酯提取物对人乳腺癌细胞SKBR-3有显著地体外增殖抑制作用,其IC50为419.0±12.87μg/ml;转基因小鼠动物模型中,金花茶二氯甲烷乙酸乙酯提取物对小鼠肿瘤的发生和发展均有显著地抑制作用;模型组小鼠肿瘤平均发生数量为5.25个/只,给药组为3.75个/只;模型组组小鼠平均瘤重为0.47g/只,给药组为0.24g/只。结论:金花茶二氯甲烷乙酸乙酯提取物对MMTV-PyMT转基因小鼠乳腺癌的发生有一定的预防作用。 |

Objective To investigate the effect of Chemical Constituents of Camelllia nitidissima Tuyama from methylene chloride ethyl acetate(JHC)on protection against tumor formation in MMTV-PyMT transgenic mice. Methods MTT assay was performed to evaluate the growth inhibitory rate of SKBR-3; The MMTV-PyMT transgenic mice model was performed to observe the protection effect in carcinogenesis of JHC in vivo. Results JHC showed significantly inhibition effect on the proliferation of SKBR-3 and the IC50 419.0±12.87μg/ml. In transgenic mice model, JHC shows significantly inhibitory effect in tumor formation and development; The average tumor number of model group is 5.25/mice, and the JHC treatment group is 3.75/mice; The average tumor weight of model group is 0.47g/mice, and the JHC treatment group is 0.24g/mice. Conclusion Chemical Constituents of Camelllia nitidissima Tuyama from Methylene chloride ethyl acetate can protect against the tumor formation in MMTV-PyMT transgenic mice. |
