      摘要 目的:分析胶质瘤组织中miR-195、miR-3653表达及其与胶质瘤临床病理特征及预后的关系。方法:于2016年1月至2019年6月采集广西医科大学第七附属医院经手术切除的42例胶质瘤患者的癌组织石蜡标本作为胶质瘤组,另选取同期行手术治疗的颅脑损伤患者30例,将术中切除的额叶或颞叶组织石蜡标本作为对照组。应用实时荧光定量聚合酶链反应 (qRT-PCR)检测胶质瘤组和对照组中miR-195、miR-3653相对表达量,并分析miR-195、miR-3653表达与胶质瘤临床病理特征的关系。Kaplan-Meier生存曲线分析miR-195、miR-3653表达与胶质瘤患者预后的关系,多因素Cox回归分析影响胶质瘤患者预后的因素。结果:胶质瘤组miR-195、miR-3653表达水平低于对照组(P<0.001)。不同胶质瘤病理分级、是否发生远处转移患者miR-195表达差异具有统计学意义(P<0.001),不同肿瘤直径、病理分级、是否发生远处转移的患者miR-3653表达差异具有统计学意义(P<0.001)。Kaplan-Meier生存曲线显示:miR-195、miR-3653低表达组3年生存率低于miR-195、miR-3653高表达组(P<0.05)。多因素Cox回归分析显示远处转移、病理分级Ⅲ~Ⅳ级、低miR-195表达、低miR-3653表达是胶质瘤患者预后的危险因素(P<0.05)。结论:胶质瘤患者miR-195、miR-3653低表达,与胶质瘤远处转移、病理分级Ⅲ~Ⅳ级以及低生存率有关,miR-195、miR-3653可能成为胶质瘤预后评估指标以及治疗靶点。

      ABSTRACT Objective: To investigate the expression of miR-195 and miR-3653 in glioma tissue and to analyze their relationship with clinicopathological features and prognosis of glioma. Methods: Collected paraffin specimens of 42 glioma patients who underwent surgical resection at The Seventh Affiliated Hospital of Guangxi Medical University from January 2016 to June 2019 as the glioma group, and another 30 patients with traumatic brain injury who underwent surgical treatment during the same period were selected, and the paraffin specimens of the frontal or temporal lobe tissue removed during surgery were used as the control group. The relative expression of miR-195 and miR-3653 in glioma group and control group were detected by real-time fluorescent quantitative polymerization chain reaction (qRT-PCR), and the relationship between the expression of miR-195 and miR-3653 and the clinicopathological characteristics of glioma were analyzed. The relationship between miR-195, miR-3653 expression and prognosis in glioma patients was analysised by Kaplan Meier survival curve. The factors affecting the prognosis of glioma patients were analysised by Multivariate Cox regression. Results: The expression level of miR-195 and miR-3653 in glioma group were lower than those in control group (P<0.001). There were significant differences in miR-195 expression among patients with different pathological grades and distant metastasis (P<0.001). There were significant differences in miR-3653 expression among patients with different tumor diameter, pathological grade and distant metastasis (P<0.001). Kaplan-Meier survival curve showed that the three years survival rate of miR-195 and miR-3653 low expression group were lower than those of miR-195 and miR-3653 high expression group (P<0.05). Multivariate Cox regression analysis showed that distant metastasis, pathological grade III-IV level, low miR-195 expression, and low miR-3653 expression were risk factors for the prognosis of glioma patients (P<0.05). Conclusion: The low expression of miR-195 and miR-3653 in glioma patients is associated with distant metastasis, pathological grading of grade III-IV level, and low survival rate, miR-195 and miR-3653 may become prognostic indicators and therapeutic targets for glioma.
