      目的 探讨长链非编码核糖核酸(LncRNA)细胞因子信号转导抑制因子2-反义转录本1(SOCS2-AS1)、微小核糖核酸(miRNA)-454-3p在人胃癌组织中的表达及临床意义。方法 选取120例行胃癌根治术患者的胃癌组织和癌旁组织。检测并对比胃癌组织和癌旁组织中LncRNA SOCS2-AS1、miR-454-3p表达,采用Pearson法分析胃癌组织中LncRNA SOCS2-AS1与miR-454-3p的表达相关性。随访3年,根据胃癌组织中LncRNA SOCS2-AS1、miR-454-3p表达均值将胃癌患者分为LncRNA SOCS2-AS1、miR-454-3p高/低表达组,使用Kaplan-Meier法绘制生存曲线,采用多因素COX回归分析胃癌患者预后不良的影响因素。结果 胃癌组织中LncRNA SOCS2-AS1、miR-454-3p表达均低于癌旁组织(P<0.05)。LncRNA SOCS2-AS1、miR-454-3p表达与分化程度、浸润深度、淋巴结转移、TNM分期有关(P<0.05)。胃癌组织中LncRNA SOCS2-AS1与miR-454-3p表达呈正相关(P<0.05)。LncRNA SOCS2-AS1低表达组3年总生存率(OS)低于LncRNA SOCS2-AS1高表达组,miR-454-3p低表达组3年OS低于miR-454-3p高表达组(P<0.05)。发生淋巴结转移、TNM分期Ⅲ期是胃癌患者预后不良的危险因素(P<0.05),miR-454-3p、LncRNA SOCS2-AS1高表达是胃癌患者预后不良的保护因素(P<0.05)。结论 胃癌组织中LncRNA SOCS2-AS1、miR-454-3p呈异常低表达,二者与胃癌患者病理特征及预后不良密切相关。

      Objective To investigate the expression and clinical significance of long non-coding ribonucleic acids (LncRNA) cytokine signal transduction inhibitor 2-antisense transcript 1 (SOCS2-AS1) and micribonucleic acid (miRNA)-454-3p in human gastric cancer tissues. Methods The gastric cancer tissues and adjacent tissues of 120 patients undergoing radical gastrectomy were selected. The expression of LncRNA SOCS2-AS1 and miR-454-3p in gastric cancer tissues and adjacent tissues was detected and compared, the correlation between LncRNA SOCS2-AS1 and miR-454-3p expression in gastric cancer tissues was analyzed by Pearson method. After 3 years of follow-up, gastric cancer patients were divided into high/low expression groups of LncRNA SOCS2-AS1 and miR-454-3p according to the mean expression of LncRNA SOCS2-AS1 and miR-454-3p in gastric cancer tissues, the survival curve was drawn by Kaplan-Meier method, and the influencing factors of poor prognosis in gastric cancer patients were analyzed by multivariate COX regression. Results The expression of LncRNA SOCS2-AS1 and miR-454-3p in gastric cancer tissues was lower than that in adjacent tissues (P<0.05). The expression of LncRNA SOCS2-AS1 and miR-454-3p was related to the degree of differentiation, depth of invasion, lymph node metastasis and TNM stage (P<0.05). There was a positive correlation between the expression of LncRNA SOCS2-AS1 and miR-454-3p in gastric cancer tissues (P<0.05). The 3-year overall survival (OS) in LncRNA SOCS2-AS1 low expression group was lower than that in LncRNA SOCS2-AS1 high expression group, and the 3-year OS in miR-454-3p low expression group was lower than that in miR-454-3p high expression group (P<0.05). Lymph node metastasis and TNM stage III were risk factors for poor prognosis in gastric cancer patients (P<0.05). High expression of miR-454-3p and LncRNA SOCS2-AS1 was a protective factor for poor prognosis in gastric cancer patients (P<0.05). Conclusion The abnormal low expression of LncRNA SOCS2-AS1 and miR-454-3p in gastric cancer tissues is closely related to the pathological characteristics and poor prognosis of gastric cancer patients.
