摘要 目的:建立高效液相色谱(HPLC)-电喷雾检测器(CAD)法测定两性霉素B脂质体(L-AMB)中氢化大豆磷脂酰胆碱(HSPC)和磷脂二硬酯酰磷脂酰甘油(DSPG)含量的方法。方法:采用Venusil XBP C 8(4.6 mm×250 mm,5 μm)色谱柱,柱温:30℃;以甲醇-水作为流动相;喷雾压力:56.4 psi,喷雾温度:55℃;梯度洗脱,流速1.0 mL?min-1。结果:L-AMB各磷脂在 5~60 mg?mL-1内与峰面积呈良好的线性关系。HSPC平均回收率为100.06%,平均RSD为0.42;DSPG平均回收率100.03%,平均RSD为0.55。按HPLC-CAD 色谱条件测定L-AMB中HSPC、 DSPG含量分别为1497.66 mg?L-1、4710.93 mg?L-1,且无溶血磷脂检出。采用HPLC-ELSD 法验证,HSPC、 DSPG含量分别为1495.28 mg?L-1、4712.71 mg?L-1, 两种方法测定HSPC、 DSPG含量基本一致。结论:HPLC-CAD 测定L-AMB中HSPC、DSPG含量操作简单、准确,可准确测定L-AMB 中脂质体的质量。 |

ABSTRACT Objective: To establish a method for the determination of hydrogenated soybean phosphatidylcholine (HSPC) and phosphatidylglycerol (DSPG) in amphotericin B liposomes (L-AMB) by HPLC and electrospray detector (CAD). Methods: Venusil XBP C 8 (4.6 mm×250 mm, 5 μm) column was used. The column temperature was 30℃. Using methanol-water as mobile phase; Spray pressure: 56.4 psi, spray temperature: 55℃; Gradient elution at flow rate of 1.0 mL?min-1. Results: L-AMB phospholipids had a good linear relationship with peak area in 5~60 mg?mL-1. The average recovery of HSPC was 100.06%, and the average RSDS were 0.42. The average recovery of DSPG was 100.03%, and the average RSDS were 0.55. HSPC and DSPG contents in L-AMB were 1497.66 mg?L-1 and 4710.93 mg?L-1, respectively, and no lysophospholipids were detected by HPLC-CAD. HSPC and DSPG contents were 1495.28 mg?L-1 and 4712.71 mg?L-1, respectively. HSPC and DSPG contents by HPLC-ELSD method were consistent. Conclusion: Determination of HSPC and DSPG in L-AMB by HPLC-CAD is simple and accurate, and can accurately determine the quality of liposome in L-AMB. |
