      摘要 目的:分析丹参酮ⅡA(TIIA)结合有氧运动(AE)对高血压大鼠血管功能的影响及作用机制。方法:选择30只自发性高血压大鼠(SHR),随机分为模型组、TⅡA组、TⅡA+AE组,每组10只,另选取10只健康大鼠作为对照组。TⅡA组大鼠给予20 mg/kg的TⅡA,TⅡA+AE组在此基础上进行中等强度的有氧运动,模型组和健康对照组大鼠均灌胃给予等体积的生理盐水溶液。观察各组大鼠收缩压(SBP)和舒张压(DBP)的变化,HE染色观察胸主动脉组织形态学变化,测定血管舒缩功能,检测血清血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)和一氧化氮(NO)的表达水平,测定胸主动脉中腺苷酸活化蛋白激酶(AMPK)、核因子E2相关因子(Nrf2)、血红素加氧酶1(HO-1)、醌氧化还原酶1(NQO-1)蛋白的相对表达。结果:TⅡA组和TⅡA+AE组大鼠的SBP和DBP均低于同期的模型组大鼠;TⅡA+AE组大鼠的SBP和DBP在第8周时均低于同期TⅡA组大鼠(P<0.05);与模型组相比,在1×10-7、1×10-6和1×10-5mol/L的PE浓度下TⅡA组和TⅡA+AE组大鼠胸主动脉血管环的收缩率均降低,且TⅡA+AE组大鼠低于同浓度下的TⅡA组(P<0.05);在1×10-8、1×10-7和1×10-6的Ach浓度下TⅡA组和TⅡA+AE组大鼠胸主动脉血管环的舒张率高于模型组,且ⅡA+AE组大鼠高于同浓度下的TⅡA组(P<0.05);TⅡA组和TⅡA+AE组大鼠血清AngⅡ和ET-1的表达水平低于模型组、NO的表达水平高于模型组(P<0.05);与TⅡA组相比,TⅡA+AE组大鼠血清AngⅡ和ET-1的表达水平较低、NO的表达水平较高(P<0.05);TⅡA组和TⅡA+AE组大鼠胸主动脉中AMPK、Nrf2、HO-1、和NQO-1蛋白的相对表达显于模型组,且TⅡA+AE组高于TⅡA组(P<0.05)。结论:TⅡA联合有氧运动可降低SHR的血压,改善血管舒缩功能和内皮功能,其机制可能与AMPK/Nrf2信号通路有关。

      ABSTRACT Objective: To investigate the effects of tanshinone ⅡA(TIIA) combined with aerobic exercise(AE) on vascular functions of hypertensive rats and its mechanism. Methods: 30 spontaneously hypertensive rats (SHR) were selected and randomly divided into the model group, TA group, and TA + AE group, with 10 rats in each group, and another 10 healthy rats were selected as the control group. Rats in the TA group received 20 mg / kg TA, in the TA + AE group performed moderate intensity aerobic exercise on this basis, and rats in the model group and healthy control group received equal volume of saline solution by gavage. The changes of systolic blood pressure (SBP) and DBP), morphological changes of thoracic aorta, vasomotor function, angiotensin serum angiotensin (Ang), endothelin-1 (ET-1) and nitric oxide (NO) were measured, and the relative expression of adenylate-activated protein kinase (AMPK), nuclear factor E2-related factor (Nrf 2), heme oxygenase 1 (HO-1) and quinone oxidoreductase 1 (NQO-1) protein in thoracic aorta was determined. Results: The SBP and DBP of TIIA group and TIIA+AE group were lower than model group; and TIIA+AE group was lower than TIIA group at 8th week(P<0.05). Compared with the model group, the rate of constriction decreased in rats at 1×10-7, 1×10-6 and 1×10-5 mol/L, and rats with TA + AE group were lower than in the TA group at the same concentration (P<0.05). The relaxation rate was higher in the thoracic aorta of the rats at 1×10-8, 1×10-7 and 1×10-6 Ach concentrations than in the model group, and the rats with A + AE were higher than in the TA group at the same concentration (P<0.05). The expression levels of serum Ang and ET-1 in TA and TA + AE rats were lower than those in the model group, and the NO expression levels were higher than those in the model group (P<0.05). Compared with the TA group, serum Ang and ET-1 in the TA + AE group were lower (P<0.05). The relative expression of AMPK, Nrf 2, HO-1, and NQO-1 proteins in the thoracic aorta of the TA and TA + AE groups was higher in the model group and higher in the TA + AE group than in the TA group (P<0.05). Conclusion: TIIA combined with AE can significantly decrease the blood pressure of SHR, ameliorate the vascular contraction and diastolic functions and endothelial functions, the potential mechanism is related with AMPK/Nrf2 signal pathways.
