      目的:研究不同烟熏强度对SD 大鼠骨骼肌组织抗氧化能力及氧化应激损伤的影响,探讨COPD骨骼肌功能障碍的机制。方 法:40 只SD大鼠随机分为对照组、实验1 组、实验2 组、实验3 组,每组各10 只。实验1 组、实验2 组、实验3 组分别给予4 周、 12 周、20 周的烟熏强度,烟熏结束后取大鼠双侧伸趾长肌,用酶联免疫吸附测定法(ELISA)方法检测各组大鼠的伸趾长肌组织匀 浆谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)、谷胱甘肽过氧化物酶(GPx)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和丙二醛 (MDA)的浓度,伸趾长肌组织蛋白质提取液羰基化蛋白(PC)的浓度、伸趾长肌组织DNA 中8- 羟基脱氧鸟苷(8-OHdG)的含量。 结果:与对照组相比,实验1 组、实验2 组、实验3 组GSH、GSH/ GSSG、SOD显著降低(P<0.05~P<0.01),GSSG 显著升高(P< 0.05),实验1 组GPx显著降低(P<0.05),实验1 组CAT显著升高(P<0.05),实验2 组、实验3 组MDA、8-OHdG 显著增高(P< 0.05~P<0.01),实验3 组PC 显著增高(P<0.05)。结论:烟熏可使大鼠骨骼肌组织抗氧化/氧化失衡,导致氧化应激损伤,并与烟 熏强度相关。

      Objective:To study the effect of anti-oxidants and oxygen stress damage on skeletal muscle in rats with different intensities of passive cigarette smoking and to explore mechanisms of skeletal muscle dysfunction in COPD.Methods:Forty rats were divided into 4 groups randomly: control group, experiment 1 group, trial 2 and 3 group. Rats of trial 1, 2, 3 groups were exposed passive cigarette smoking for 4 weeks, 12 weeks and 20 weeks respectively. GSH, GSSG, GPx, CAT, SOD and MDA, PC, 8-OHdG in the extensor digitorum longus of rats were detected through ELISA.Results:The expression levels of GSH, GSH/GSSG, SOD decreased and GSSG increased significantly in all trial groups (P<0.05~P<0.01) , GPx decreased and CAT increased significantly in trial 1 group (P<0.05) , and MDA, 8-OHdG in trial 2, 3 groups and PC in trial 3 group increased significantly in comparison with control group (P<0.05~P< 0.01).Conclusion:Cigarette smoking can cause imbalance of anti-oxidants /oxygen stress and results in oxygen stress damage in the distal skeletal muscle tissue.
