      目的:探讨大鼠脂肪来源干细胞(adipose-derived stemcells,ADSCs)对放射后成纤维细胞(fibroblast,Fb)的影响。方法:体外原 代培养SD大鼠ADSCs 和真皮Fb,分别对两种细胞进行鉴定,Fb 辐照后两种细胞建立共培养模型。将Fb分为3 组,对其进行 lncRNA 和mRNA转录本高通量测序。结果:ADSCs 高表达CD29 和CD44,低表达CD31 和CD45。大鼠Fb 高表达波形蛋白。Fb 单纯辐照和干细胞干预后转录水平有明显的差异。结论:ADSCs 能使辐射后的Fb 细胞lncRNA 和mRNA转录水平明显的改变, 提示多项生物学功能及信号通路在其中发挥作用。

      Objective:To discuss the effect of rat adipose derived stem cells (ADSCs) on fibroblasts (Fb) after radiation.Methods:We cultivated ADSCs and dermal Fb of SD rats in vitro, and two kinds of cells were identified. Then a co-culture model was established. Fb was divided into 3 groups, which carry on lncRNA and mRNA high throughput sequencing.Results:The high expression of CD29 and CD44 were identified in ADSCs, and the low expression of CD31 and CD45 in Fb. There was a significant difference in the level of transcripts between control group and Fb group of ADSCs Intervention after radiation.Conclusion:ADSCs can significantly alter the lncRNA and mRNA transcription levels of Fb cells after irradiation, suggesting that a number of biological functions and signaling pathways play an important role in the prcess.
