      目的:探讨miR-155 对前列腺癌细胞周期的影响及其分子机制。方法:通过转染anti-miR-155 抑制前列腺癌DU145 和PC-3 细胞中miR-155 水平后,采用流式细胞术观察细胞周期的变化,western blot 和RT-PCR 观察p53 和p21蛋白及CDK2 和cyclin 蛋白和mRNA表达的变化。结果:与对照组相比,DU145 和PC-3 细胞转染anti-miR-155 后,G2/M期细胞阻滞,S期细胞数比例显 著增加(P<0.05),p53 和p21 蛋白和mRNA表达水平显著增加(P<0.01),CDK2 和cyclin E蛋白和mRNA 表达均显著降低(P<0. 01)。结论:miR-155 可影响人前列腺癌细胞的周期,可能与其调节p53、p21 及其下游的CDK2 和cyclin E的表达相关。
英文摘要:
      Objective:To study the effects of miR-155 on the cell cycle of prostate cancer and the underlying mechanisms.Methods:The cell cycles of DU145 and PC-3 cells were evaluated after anti-miR-155 transfection by FACS. p53 and p21 expression were evaluated by western blot and RT-PCR.Results:The cell cycle of DU145 and PC-3 cells were down regulated obviously after anti-miR-155 transfection(P<0.05), and the protein and mRNA expression of p53 and p21 were obviously increased (P<0.01). In addition, the protein and mRNA expression of CDK2 and cyclin E were also decreased obviously (P<0.01).Conclusion:Inhibition of miR-155 could modulate the cell cycles of DU145 and PC-3 cells in vitro by p53/p21 pathways and by down regulating the expression of CDK2 and cyclin E.
