Objective:To explore the relationship between CDH1 gene promoter methylation and clinical pathological types of
cervical cancer.Methods:105 cases of specimens from patients with cervical cancer (Cervical cancers group) and 60 cases of normal
cervical (Normal controls group) were selected, then methylation specific polymerase chain reaction (MSP) was used to detect the CDH1
gene start promoter CpG island methylation status of the samples and the high-risk HPV DNA status. After that we analyzed the
relationship between CDH1 gene methylation status and high-risk HPV DNA status, CDH1 gene methylation status and clinical
parameters of patients.Results:The positive rate of CDH1 gene promoter methylation in cervical cancer group was 56.19%, significantly
higher than that in the normal group (6.67%), with statistical difference (P<0.05). The positive rate of DNA HPV in cervical cancer group
was 84.76%, which was significantly higher than that in the normal group (20%), and the difference was statistically significant (P<0.05).
The consistency analysis results of DNA HPV and CDH1 gene promoter methylation were statistically significant (P<0.05). CDH1 gene
promoter methylation rate was significantly associated with the degree of WHO differentiation, FIGO stage, histological grade, tumor
size, which was statistically significant (P<0.05).Conclusion:The methylation of CDH1 gene promoter in cervical cancer is associated
with many clinical parameters (WHO differentiation grade, FIGO grades, Histological types, Tumor diameter), and is consistent with the
high risk type DNA HPV positive. It can be used as a reference index for the diagnosis and prognosis of cervical cancer. |
