| 目的：观察肿瘤坏死因子样弱凋亡诱导因子(TWEAK)对人肝星状细胞(HSCs)株LX-2 增殖、细胞周期、细胞凋亡、迁移及粘
附能力的影响。方法：用不同浓度TWEAK培养LX-2 细胞24 或48 h，采用CCK-8法检测TWEAK对LX-2 细胞增殖的影响，流
式细胞术检测TWEAK对LX-2 细胞周期和凋亡的影响，Transwell 小室检测TWEAK 对LX-2 细胞迁移能力的影响，Matrigel 基
质胶检测TWEAK 对LX-2细胞的粘附能力的影响。结果：与对照组相比，40 ng/mL和100 ng/mL TWEAK 都能使LX-2细胞的迁
移能力增强(P<0.05)，且100 ng/mL较40 ng/mL作用更强(P<0.01)；100 ng/mL TWEAK 能够明显抑制LX-2 细胞的粘附能力(P<0.
0001)；40 ng/mL、100 ng/mL TWEAK 对LX-2 细胞的增殖、周期及凋亡无明显影响(P>0.05)。结论：TWEAK 能够增强LX-2 细胞迁

| Objective:To explore the effects of tumor necrosis factor-like weak inducer of apoptosis (TWEAK) on proliferation,
migration, adhesion, cell cycle and apoptosis of human hepatic stellate cells (HSCs) line LX-2.Methods:LX-2 was incubated with
different concentrations of TWEAK for 24 h or 48 h, and the effects of TWEAK on the proliferation of LX-2 was investigated by Cell
Counting Kit-8 (CCK-8). The flow cytometry was used to detect the change of cell cycles and apoptosis, and Transwell assay to detect
the migration, Matrigel to evaluate adhesion.Results:Compared with controls, 40 ng/mL and 100 ng/mL TWEAK could promote LX-2
migration (P<0.05), and 100 ng/mL had a stronger effect (P<0.01); 100 ng/mL TWEAK inhibited cell adhesion (P<0.0001); The
proliferation, cell cycles and apoptosis of LX-2 was not influenced by different concentrations of TWEAK (P>0.05).Conclusion:TWEAK could promote the migration and inhibit the adhesion of LX-2 cells. |
