      目的:本研究探索神农架大龙潭川金丝猴TRIM5alpha 蛋白序列的特征、变异、该基因进化特点以及该基因的遗传多样性,对 金丝猴的保护和研究具有重要的意义。方法:从神农架川金丝猴投食群随机选择41 个个体,采集毛发并提取基因组DNA,设计引 物,对其TRIM5 alpha 基因编码序列进行PCR 扩增和测序,最后对序列进行比对和分析。结果:在神农架川金丝猴投食群41 个个 体中,TRIM5alpha 基因不存在与CypA 假基因序列融合的现象。与恒河猴序列不同而与人类相似的是,川金丝猴中没有发现 TFP339-341 型,而只发现△△Q339-341 型。41 个个体TRIM5alpha 蛋白序列没发现任何非同义突变位点,显示该基因在该种群中 具有很低的遗传多样性。编码序列的进化分析表明,Coiled-coil 结构域和SPRY 结构域在进化中经历了明显的正向选择,Ring 结 构域和B-box 结构域则经历了纯化选择。结论:川金丝猴TRIM5 alpha 基因存在很多独特变异位点,且经历了明显的正向选择。对 投食群的研究表明该基因编码序列在该种群中多样性很低,这从侧面反映了该总群较低的遗传多样性。针对这种情况,建议在种 群复壮的工作中,适当引入一批有效的建群者。

      Objective:In this investigation, we explore the sequence characteristics, mutation, gene evolution characteristics and the genetic diversity of TRIM5 alpha sequence in golden snub-nosed monkey provisioned group in Shennongjia National Natural Reserve.Methods:We selected 41 individuals randomly from the population, collected hair samples and extracted the genomic DNA. Then we designed the primers for PCR amplification taking rhesus monkeys TRIM5 alpha gene sequence as a reference and the products was sequenced. Finally, we alignment and analysis the sequence.Results:We did not detected a gene fusion phenomenon of TRIM5alpha and CypA pseudogene sequences in the 41 individuals. Compared with rhesus monkeys protein sequences, there is no genotype TFP339-341 was found, there is only genotype△△Q339-341 instead, which is similar to the TRIM5 alpha sequence of human. Sequence alignment of TRIM5alpha protein did not detected any non-synonymous mutations, showed that the gene has very low genetic diversity in the population. The analysis for the evolution of coding sequence of the gene shows that the Coiled-coil domain and the SPRY domain have been experiencing positive selection obviously in the evolution, the Ring domain and B-box domain have been experiencing purification selection.Conclusion:There is much unique mutation loci in the golden snub-nosed monkey TRIM5 alpha gene and this gene experienced obvious positive selection in the evolution. Studies on TRIM5 alpha in Golden Snub-nosed Monkey Provisioned Group in Shennongjia National Natural Reserve have shown that the gene coding sequence in the population diversity is rather low, suggested in fellowing work of population rejuvenation, the group needs to recruit suitable individuals to adapt the environment.
