Objective:To discuss the protective effects of different doses of Leptin pretreatment on the Myocardial Ischemia
Reperfusion Injury in Diabetic Rats.Methods:According to the random number table, 72 rats were divided into normal control group,
sham operation group, myocardial infarction group and large, medium, small dose of Leptin pretreatment groups. Normal control group
were fed with normal diet, the other groups were fed with high sugar and high fat feeding. After 1 months, all rats were abdominal vein
blood, and then the FBG, FTNS, leptin, HOMA, TNF-α, MDA and serum lipid levels were measured in rats.Results:Compared with the
control group, the FBG, fins, leptin level and HOMA-IR of other group were significantly rised (P<0.05); Compared with false-operation
group, myocardial infarction group and large, medium, small dose of Leptin pretreatment group rats FBG and leptin levels were
significantly increased, myocardial infarction group and high dose group rats, fins and HOMA-IR were significantly increased (P<0.05),
low, middle dose of Leptin pretreatment group rats, fins and HOMA-IR were significantly lower (P<0.05); Compared with myocardial
infarction group, the levels of FBG, FINS, leptin and HOMA-IR in small, medium dose of Leptin pretreatment group were significantly
lower (P<0.05). Compared with the control group, the TNF-琢, MDA levels of other groups were significantly increased (P<0.05);
Compared with false-operation group, myocardial infarction group, myocardial infarction group and large, medium, small dose of Leptin
pretreatment group rats TNF alpha, MDA level increased significantly (P<0.05); Compared with the myocardial infarction group, small
dose of Leptin pretreatment group rats TNF alpha, MDA level were significantly lower (P<0.05). Compared with the control group, the
TC, TG, LDL-C levels of other groups were significantly increased, HDL-C levels were significantly reduced (P<0.05); Compared with
the shamoperation group and large, medium, small dose of Leptin pretreatment group rats HDL-C levels were elevated, and levels of TG,
TC, LDL-C were significantly lower (P<0.05); compared with the myocardial infarction group, small dose of Leptin pretreatment group
rats TG, TC, LDL-C levels were significantly lower (P<0.05), HDL-C levels were significantly increased (P<0.05).Conclusion:Leptin
pretreatment can protect myocardial ischemia and reperfusion injury in diabetic rats, which may be related to the reduction of inflammation, oxidative stress, blood lipid disorder and insulin resistance. |
