      目的:探讨利拉鲁肽对载脂蛋白E 基因缺陷(ApoE-/-)小鼠动脉粥样硬化(AS)的影响。方法:将24 只无特定病原体(SPF)级 雄性ApoE-/-小鼠(6 周龄)随机分成对照组与实验组,各12 只,分别用高脂饲料、高脂饲料+ 利拉鲁肽饲养,饲养12 周后,比较 两组小鼠AS 斑块面积/管腔面积、内膜厚度/中膜厚度及血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高 密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、肿瘤坏死因子琢(TNF-琢)及丙二醛(MDA)水平。结果:实验组AS斑块面积/ 管腔面 积和内膜厚度/中膜厚度均明显低于对照组(P<0.05)。实验组血清TC、TG及LDL-C 水平均明显低于对照组(P<0.05),但两组小 鼠血清HDL-C 水平比较差异无统计学意义(P>0.05)。实验组小鼠血清NO 水平明显高于对照组、血清TNF-α水平明显低于对照 组(P<0.05),但两组小鼠血清MDA水平比较差异无统计学意义(P>0.05)。结论:利拉鲁肽能够改善ApoE-/- 小鼠血脂水平、保护 细胞内皮功能、抑制炎症反应,抗AS 作用显著。

      Objective:To study the influence of Liraglutide on atherosclerosis (AS)in apolipoprotein E deficient (ApoE-/-) mice.Methods:A total of 24 specific pathogen free (SPF)male ApoE-/- mice were divided randomly into control group and experiment group, with 12 cases in each group, respectively fed with high fat diet and high fat diet+Liraglutide, after 12 weeks feeding,compared the plaque area/lumen area and intima thickness/intima-media thickness of AS, as well as the levels of serum total cholesterol(TC), triglyceride(TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), nitric oxide (NO), tumor necrosis factor-α (TNF-α)and malondialdehyde (MDA).Results:The plaque area/lumen area and intima thickness/intima-media thickness of AS in experiment group were significantly lower than control group(P<0.05).The levels of serumTC, TG and LDL-C in experiment group were significantly lower than control group (P<0.05), but there was no significantly differences of the level of serumHDL-C of the two groups (P>0.05). The level of serum NO in experiment group was significantly higher than control group, on the contrary, the level of serum TNF-αin experiment group was significantly lower(P<0.05), but there was no significantly differences of the level of serum MDA of the two groups(P>0.05).Conclusion:Liraglutide can obviously improve the levels of serumlipid, protect cell endothelial function and inhibit inflammatory reaction of ApoE-/- mice, which effect on anti AS is significant.
