位液压损伤装置建立大鼠颅脑损伤(TBI)模型,术前及术后给予厄贝沙坦治疗,用激光多普勒测定局部脑区血流(rCBF)的变化,
术前及术后1、3、5 和7d 利用神经功能评分评估大鼠神经功能损伤,利用TUNEL 染色检测大鼠脑细胞凋亡情况,利用Western
Blot检测大鼠脑组织损伤周围区域活性caspase 3的表达。结果:与正常值相比,TBI手术后损伤局部脑区rCBF下降至30 %(P<0.

Objective:To investigate the neuroprotective effects of irbesartan on after fluid percussion-induced brain injury in rats.Methods:Traumatic brain injuries (TBI) model was prepared usingfluid percussion injury method and treated with irbesartan. Regional
cerebral blood flow (rCBF) and neurological status was evaluated at different time point respectively. Apoptosis was evaluated by
TdT-mediated dUTP nick end labeling (TUNEL) staining and active caspase 3 was detected.Results:Compared with sham group rats,
the rCBF was decreased to 30%in TBI rats (P<0.05). The neurological score was decreased significantly (P<0.05). TUNEL positive cells
and expression of active caspase 3 were increased in the peri-infarct cortex (P<0.05). Blockade of brain (angiotensin Ⅱ type 1)AT1
receptors with irbesartan before TBI could increase rCBF and reduce the infarct size on day 7 after TBI and increased neurologicalstatus
(P<0.05). TUNEL positive cells and expression of active caspase 3 were decreased in TBI rats after irbesartan treatment (P<0.05).Conclusion:Irbesartan can play neuroprotective effects by inhibiting apoptosis. |
