      目的:探讨内质网应激分子葡萄糖调节蛋白78(GRP78)对非小细胞肺癌(NSCLC)的诊断价值。方法:收集45 例NSCLC 患 者术后癌组织标本,及其18 例癌旁组织,采用实时定量PCR法检测所有组织中GRP78 的表达情况,分析NSCLC 癌组织中 GRP78 的表达与患者临床特征的关系,及其对患者生存期的影响。结果:NSCLC 组织中GRP78 高表达率明显高于癌旁组织,差 异有统计学意义(P<0.05)。而Ⅲ a 期NSCLC 患者癌组织中GRP78 的表达水平明显高于Ⅰ-Ⅱ期,两者存在显著性的差异(P<0. 05)。Kaplan-Meier 分析显示,GRP78 高表达的患者其生存期明显低于GRP78 低表达的患者(P<0.05)。结论:NSCLC 患者GRP78 的表达可能与肿瘤细胞的发生、发展及患者的生存期有关,可作为一个预测NSCLC 患者诊断及治疗的重要的分子标志物。
英文摘要:
      Objective:To explore the diagnostic value of glucose regulated protein 78 (GRP78) of endoplasmic reticulum stress molecules for non small cell lung cancer (NSCLC).Methods:A total of postoperative carcinoma tissue specimens of 45 patients with NSCLC were selected, fromwhich 18 cases of adjacent to carcinoma tissues were selected at the same time. The expressions was used to of GRP78 of all the selected tissues were detected by Real-time PCR and analyzed the correlation of the expression of GRP78 in NSCLC tissues with clinical characteristics and the survival periods of the patients.Results:The expression levels of GRP78 in NSCLC tissues were much higher than those in para-cancerous tissues, the differences were statistically significant (P<0.05). The expression levels of GRP78 in the cancer tissues of patients with NSCLC at stage Ⅲ a were much higher than those at stage I-II, the differences were statistically significant (P<0.05). Kaplan-Meier analysis showed that the survival periods of patients with high expression level of GRP78 were shorter than low expression level of GRP78(P<0.05).Conclusion:The expression of GRP78 May be related to the occurrence and development of tumor cells and the survival periods of patients,which can be used as an important biomarker of forecasting diagnosis and treatment of NSCLC patients.
