目的:制备担载血管生长因子(VEGF)的乳液法静电纺丝纤维膜,对其开展一系列表征,从而研究其血管再生的潜能。方法:
通过W/O 乳液法制备担载VEGF的静电纺丝纤维膜,并对其形态、力学性质进行表征。用VEGF ELISA 分析方法对其体外释放
动力学进行研究。运用CCK-8 法检测乳液法静电纺丝纤维膜中VEGF的活性变化。结果:乳液法静电纺丝纤维膜呈现连通的三维
网状结构,平均直径为1 滋m,模拟了细胞外基质(ECM),最大拉伸应力为3.03± 0.66 MPa,具有良好的抗拉伸能力,能够支持细胞
的生长。乳液法纤维膜中VEGF在体外累积释放了14天,总释放量超过20000 pg,达到血管再生的有效浓度。CCK-8 结果显示,
乳液法纤维膜中的VEGF仍然保持较高的蛋白活性。结论:担载VEGF的乳液法静电纺丝纤维膜能够缓释出活性的蛋白,具有血
管再生的潜能。 |

Objective:To study the potential of angiogenesis by preparing emulsion method electrospun fibers loading vascular
endothelial growth factor (VEGF) and carrying out a series of characterization.Methods:Electrospun fibers loading VEGF were prepared
by W/O emulsion method and were characterized in morphology and mechanical properties. The release kinetic of VEGF in electrospun
fibers was detected by ELISA method in vitro. By CCK-8 method, the changes of active situation of VEGF in electrospun fibers were
investigated.Results:Emulsion method electrospun fibers were connected by three-dimensional network structure with average diameter
1 滋m, which simulated the extracellular matrix (ECM) and they had good tensile strength which could support cell growth, their
maximum tensile stress was 3.03± 0.66 MPa. VEGF in emulsion method fibers had cumulative release of 14 days, the total amount was
over 20000 pg which could reach the effective concentration of angiogenesis. The result of CCK-8 tests showed that VEGF loaded in
emulsion method fibers still maintained high activity.Conclusion:Emulsion method electrospun fibers loading VEGF had a
sustained-release of active protein, which had the potential of angiogenesis. |
