目的:探讨血管紧张素Ⅱ(AngⅡ)皮下灌泵诱导Apo E-/-小鼠建立腹主动脉瘤模型的特点和可行性。方法:选取8-10 周龄雄
性Apo E-/-小鼠,通过植入式微量渗透释放泵皮下泵入Ang Ⅱ(1000 ng/kg·min),持续泵入28 天;对照组小鼠泵入生理盐水28 天。
取材后,测量小鼠腹主动脉直径,固定包埋做石蜡切片进行HE 及弹力纤维染色,用Real-time PCR 方法检测腹主动脉中α- 平滑
肌肌动蛋白(α-SMA)和骨桥蛋白(OPN)的表达。结果:AngⅡ处理组小鼠成瘤率为80%,腹主动脉直径较生理盐水对照组明显增加
(P<0.05);病理染色显示炎症细胞明显增多,弹力纤维断裂增多,胶原合成增多。小鼠腹主动脉琢-SMA mRNA表达较对照组显著
降低(P<0.05),而OPN mRNA表达较对照组明显增高(P<0.05)。结论:通过AngⅡ连续灌注28 天可以诱导Apo E-/-小鼠发生腹主
动脉瘤,腹主动脉平滑肌细胞由收缩型向分泌型转化可能作为小鼠腹主动脉瘤发生的鉴定及治疗靶点之一。 |

Objective:To investigate the feature and feasibility of establishing a mouse model of abdominal aortic aneurysm induced
by Ang II infusion into Apo E-/- mice.Methods:Apo E-/- mice aged 8-10 weeks were subcutaneous infused with angiotensin Ⅱ
through an implantable osmotic pump for 28 days at a rate of 1000 ng/kg·min. The control group was infused with saline. The abdominal
aorta was observed in vivo after infusion for 28 days. At last, the pathological changes of the abdominal aorta were compared with the
control group, including the abdominal aortic diameter, morphological changes of the aortic wall with HE staining, collagen content in
the vascular wall with masson staining and elastin content with elastin staining. The expression of OPN mRNA and α-SMA mRNA was
detected with real-time PCR.Results:The tumor rate in Apo E-/- mice infused with Ang II was 80%, and the abdominal aortic diameter of
AngⅡ group was markedly increased compared with the control group (P<0.05). Pathological staining revealed a significant increase of
inflammatory cells and increased size and sumof smooth muscle cells. Elastin staining showed that elastic fibers were disrupted in AngⅡ
group. Masson staing showed that collagen content was increased after AngII treatment. Real-time PCR results showed that α-SMA mRNA
was down-regulated(P<0.05) while OPN mRNA was up-regulated after Ang II treatment(P<0.05).Conclusion:Abdominal aortic
aneurysmmodel could be successfully established with 1000 ng/ kg·min of angiotensin Ⅱfor 28 days subcutaneous infusion into Apo E-/-
mice. The phenotype transformation in smooth muscle cell might be a factor for the diagnose of abdominal aortic aneurysm. |
