      目的:研究RGD肽对肺癌A549细胞增殖凋亡及侵袭迁移的影响,并探讨其作用机制。方法:不同浓度RGD 肽处理肺癌 A549 细胞后,MTT 检测肺癌细胞的增殖能力,流式细胞仪检测肺癌细胞凋亡及周期分布,Transwell 检测其迁移及侵袭能力的变 化,Western blot 检测RGD肽对肺癌A549 细胞MMP2、MMP9的表达水平影响。结果:当RGD 肽浓度增加至50 mg/L 时,肺癌 A549 细胞增殖明显受到抑制,且这种抑制作用呈剂量依赖关系;RGD肽组A549 细胞G0/G1 期细胞比例增高,细胞凋亡率由 (6.1± 0.1)%增至(15.2± 0.5)%;在迁移和侵袭试验中,RGD肽组A549细胞的穿膜细胞数分别由123± 10 和43± 10 降至45± 5 和18± 5;RGD肽组A549 细胞MMP2、MMP9表达水平显著降低。结论:RGD肽对肺癌A549细胞的增殖有明显抑制作用,并 促进其凋亡,可能与RGD 肽改变其周期分布有关,RGD 肽可明显抑制A549 细胞的迁移及侵袭,可能与其下调MMP2、MMP9 的 表达相关。

      Objective:To investigate the effect of RGD peptide on the proliferation, apoptosis, migration and invasion of the lung cancer A549 and explore its mechanism.Methods:MTT was performed to reveal the inhibitory effect on cell proliferation after different doses of RGD peptide were added to the A549 cell. Flow cytometry was used to determine the apoptosis and cell cycle of A549 cell. Transwell was performed to identify the migration and invasion of A549 cell. The expression level of MMP2 and MMP9 was measured by Western blot.Results:When the concentration of RGD peptide increased to 50 mg/L, there was a dose-dependent inhibition of cell proliferation of A549. In RGD peptide group, the rate of GO/G1 increased and apoptosis rate of A549 increased from (6.1± 0.1)% to (15.2± 0.5) %. In RGD peptide group, the number of migrated and invaded cells decreased from 123± 10, 43± 10 to 45± 5, 18± 5, respectively. Moreover, RGD peptide down-regulated the expression of MMP2 and MMP9.Conclusion:RGD peptide can inhibit the proliferation and promote the apoptosis of A549 cell line through changing cell cycle, suppressing the migration and invasion by down-regulating the MMP2 and MMP9 expression.
