真核翻译起始因子3(Eukaryotic translation factor 3,eIF3)是由多个亚单位组成的复合因子,其中eIF3a 是其最大的亚单位。
很多研究表明在酵母和哺乳动物细胞中,eIF3都参与了mRNA翻译起始,并对蛋白质的合成有很好的调控作用。值得一提的是
eIF3a 通过调控一系列与肿瘤的生成、细胞周期的调控DNA 修复等过程相关的mRNA的翻译从而在肿瘤的发生、演进和干预中
发挥重要作用。此外,研究发现eIF3a 对RAF-MEK-ERK 信号通路有抑制作用。eIF3a 对蛋白质翻译的调节及其对
RAF-MEK-ERK 信号通路的影响使其有望成为肿瘤治疗的新靶点。本文将着重围绕eIF3a在肿瘤发生、演进和干预中的作用进行

Eukaryotic translation factor 3 (eIF3) is a composite factor of multiple subunits. eIF3a is the largest subunit. Many
studies show that in yeast and mammalian cells, eIF3 plays a pivotal role in translational control. eIF3a has recently been shown to play
an important role in tumors genesis, evolution and intervention by regulating translation of a subset of mRNAs which are involved in
tumorigenesis, invasion and metastasis, regulation of cell cycle progression, and DNA repair pathways. Besides, eIF3a correlates with
cancer occurrence, development, platinum-based chemotherapy sensitivity and prognosis. Meanwhile, owing to the role of eIF3a in
protein synthesis and its regulation in RAF-MEK-ERK signaling pathway, it is expected to become a new target for cancer therapy. This
article focuses on the role of eIF3a in tumors genesis, evolution and intervention. |
