      目的:观察侧脑室注射食欲素A(OXA)对大鼠胃运动的作用及作用途径。方法:选取成年雄性大鼠50只,随机分为5 组,即 生理盐水(NS)组、1 ug组、5 ug 组、10 ug 组和20 ug组。在胃窦和十二指肠植入应力传感器,记录大鼠胃肠自发性运动。通过侧脑 室注射OXA,观察胃肠环形肌运动波形的变化和持续时间。选取10 只大鼠随机分为2组,一组皮下注射NS+10 ug OXA,另一组 皮下注射阿托品+10 ug OXA,观察OXA对胃肠运动的影响。同理选取10 只大鼠分为假手术组+10 ug OXA组,迷走神经切断 组+10 ug OXA组,观察OXA 调控大鼠胃肠运动的作用途径。再选取10 只大鼠,先注射NS 再注射OXA拮抗剂SB334867,观察 内源性OXA对胃肠运动的影响。结果:侧脑室注射OXA(1-20 ug),大鼠消化间期胃和十二指肠III期收缩波消失,继而出现胃和 十二指肠餐后不规则的收缩波。OXA对餐后胃肠运动的影响可被阿托品或切断迷走神经干所阻断(P<0.05)。中枢注射选择性 OXA受体拮抗剂SB-334867 (16 ug),可增强胃消化间期III相收缩(P<0.05)。结论:中枢注射OXA可能经迷走神经胆碱能通路调 控大鼠消化间期的胃肠运动,内源性OXA可能对大鼠消化间期胃肠运动具有抑制作用。

      Objective:To observe the effect and mechanismof intracerebroventricular(icv) injection of Orexin-A on gastric motility in rats.Methods:50 adult male rats were randomly divided into five groups, namely normal saline (NS), 1 滋g, 5 滋g, 10 滋g and 20 滋g group. Two strain gauge transducers were attached on the antrumand duodenumto record circular muscle contractions. Spontaneous gastroduodenal contractions were recorded in freely moving conscious rats. Through icv injection of OXA, observed the changes in the gastrointestinal circular muscle motion wave form and duration in rats. 10 rats were randomly divided into two groups, one group with subcutaneous injection of NS + icv injection of 10 ug OXA, another group with subcutaneous injection of atropine + icv injection of 10 ug OXA, to observe the effects on gastrointestinal motility. Similarly selecting 10 rats were divided into sham group + 10 ug OXA group, vagotomy + 10 ug OXA group, which was used to elucidate the neural pathways of OXA. Then selecting 10 rats, first icv injection of NS then icv injection of OXA antagonist SB334867, observed the effect of endogenous OXA on gastrointestinal motility.Results:OXA (1-20 ug) disrupted the interdigestive phase III-like contractions and induced an irregular postprandial-like motility pattern in the stomach and duodenum. The stimulatory effect of OXA on gastroduodenal postprandial-like motility pattern was abolished by atropine and truncal vagotomy(P<0.05). Central administration (icv) of selective OXA receptor antagonist, SB-334867 (16 ug), enhanced gastric spontaneous phase III-like contractions (P<0.05).Conclusion:Central administration of OXA changes GI motor pattern from interdigestive to postprandial via the vagal cholinergic pathways. Endogenous OXA may inhibit in interdigestive GI motility in rats.
