目的:探索人参苷皂Rd 对人脑胶质瘤的作用。方法:人脑胶质瘤U251 细胞系细胞培养,不同浓度的人参皂苷Rd 处理观察
细胞形态、测定端粒酶活性以及hTERT 表达水平。结果:随着GSRd 浓度的升高,U251 细胞的生长被明显抑制,出现了细胞凋亡
和凋亡小体的现象;在经GSRd处理后U251 细胞的端粒酶活性,对照组和溶媒组类似,而GSRd药物处理24 h 后,细胞端粒酶活
性显著降低,其中20 ug/L组端粒酶活性降低极显著,P<0.01,40 ug/L和80ug/L 组端粒酶活性显著降低,P<0.05。GSRd 药物处
理48 h 后,细胞端粒酶在20、40、80 ug/L处理后,端粒酶活性降低极显著,P<0.01;20 ug/L、40 ug/L和80 ug/L 的GSRd 处理细
胞后,相比于对照和溶媒组,hTERT 基因表达水平显著降低。结论:人参皂苷Rd 能够促进人脑胶质瘤U251 细胞凋亡,对于临床
治疗脑胶质瘤有重要的意义。 |

Objective:To explore the role of ginsenosides Rd for human glioma.Methods:The human glioma cell line U251 cells
were cultured, and the cell morphology, measurement of telomerase activity and hTERT expression levels were observed by by different
concentrations of ginsenosides Rd treatment.Results:With GSRd concentration increased, the growth of U251 cells was significantly inhibited;
in the U251 cells, telomerase activity similar to the control group and the vehicle treatment group after drug treatment with GSRd
for 24h, telomerase activity was significantly reduced, while 20 ug / L group telomerase activity decreased significantly, P<0.01, telomerase
activity lowered significantly in the 40 ug / L and 80 ug / L group, P<0.05, P <0.05. GSRd drug treatment after 48 h, telomerase activity
decreased significantly in the 20, 40, 80 ug / L group, P <0.01; Compared to the control and vehicle group, hTERT gene expression
levels were significantly reduced in 20 ug / L, 40 ug / L and 80 ug / L group.Conclusion:Ginsenoside Rd can promote apoptosis in human
U251 cells, it is important clinical significance for the treatment of glioma. |
